החקירה של רמות החלבון תאית של מיני חיידקים היא בעלת חשיבות להבנת המנגנונים פתוגניים במחלות הנגרמים על ידי אורגניזמים אלה. כאן אנו מתארים הליך להפקת חלבון ממינים Burkholderia מבוססת על תמוגה מכאנית באמצעות חרוזי זכוכית בנוכחות חומצת ethylenediamine tetraacetic ופלואוריד phenylmethylsulfonyl בופר פוספט. שיטה זו יכולה לשמש למיני Burkholderia שונים, לתנאי גידול שונים, וסביר להניח מתאים לשימוש במחקרי proteomic של חיידקים אחרים. לאחר מיצוי חלבון, טכניקת proteomic דו ממדים (2-D) ג&#39;ל אלקטרופורזה מתוארת ללמוד שינויים הגלובליים בproteomes של אורגניזמים אלה. שיטה זו מורכבת של הפרדת חלבונים לפי הנקודה איזואלקטרית על ידי איזואלקטרית התמקדות בממד הראשון, ואחריו הפרדה על בסיס משקל מולקולריעל ידי ג&#39;ל אלקטרופורזה acrylamide בממד השני. הדמיה של חלבונים מופרדים מתבצעת על ידי צביעת כסף.
