세균 종의 세포 내 단백질 수준의 조사가 이들 유기체에 기인 한 질병의 발병 기전을 이해하는 데 중요하다. 여기에서 우리는 인산염 완충 생리 식염수에 에틸렌 디아민 테트라 아세트산 및 페닐 메틸 불소의 존재에 유리 구슬을 사용하여 기계적인 용해에 따라 부르크 홀데 리아 종에서 단백질을 추출하는 절차를 설명합니다. 이 방법은 다른 성장 조건을 위해, 다른 부르크 홀데 종에 사용될 수 있고, 다른 균의 프로테오믹스 연구에 사용하기위한 것으로 적당하다. 단백질 추출 후, 이차원 (2-D) 겔 전기 단백체 기술은 이러한 유기체의 프로테옴에서 해외 변화를 연구하는 기술된다. 이 방법은 분자량에 기초하여 분액하여 제 차원에서 등전위 포커싱에 의해 그들의 등전점에 따라 단백질의 분리, 이루어져두 번째 차원에서 아크릴 아마이드 겔 전기 영동에 의해. 분리 된 단백질의 시각화를 실버 염색으로 수행된다.
