Insbesondere ist ein Schlüsselfaktor in der neuronalen Verletzungen in der Schlaganfall-Kalzium Deregulierung, Parkinson, Alzheimer und anderen neurodegenerativen Erkrankungen 3,5. Daher ist es von entscheidender Bedeutung, um quantitativ zu verstehen, wie Calcium in den Zellen verteilt und wie diese Veränderungen nach physiologischen oder pathophysiologischen Stimuli. Frei in Lösung oder an ein Substrat gebunden - - und das zelluläre Calciumkonzentrationen über mehrere Größenordnungen als Folge der Stimulation verändern Dieses Ziel wird durch die Tatsache, dass Kalzium wird dynamisch zwischen zwei physikalische Zustände verteilt kompliziert. Vorteile der ESMA sind einzelne Organellen-Level-Auflösung und Empfindlichkeit submillimolaren. In der Praxis jedoch, ESMA erfordert spezielle cryotechniques und Instrumente für die Probenvorbereitung und Analyse. Hier werden die Werkzeuge, Techniken und Instrumente für die Messung von intrazellulären Kalzium mittels ESMA entsprechende beschrieben. Intramitochondrialen Kalzium ist besonders interest wegen der entscheidenden Rolle, dass die mitochondriale Calciumüberladung spielt bei neurodegenerativen Erkrankungen.

In diesem Artikel werden die Werkzeuge, Techniken und Instrumente für quantitative Messungen der intrazellulären Elementgehalt falls unter Verwendung der Technik, die als Elektronenstrahl-Mikroanalyse (ESMA) bekannt sind, beschrieben. Schockgefrieren von kultivierten Zellen oder organotypischen Kulturen wird durch Eint Einfrieren in flüssigem Ethan oder durch Einfrieren Slam gegen einen kalten Metallblock bzw. durchgeführt. Cryoschnitte nominell 80 nm dick sind trocken mit einem Diamantmesser bei ca. geschnitten. -16076, C, auf Kohlenstoff / PIOLOFORM beschichteten Kupfergitter montiert und cryotransferred in eine Kryo-EM mit Hilfe eines speziellen cryospecimen Halter. Nach der visuellen Mapping-Umfrage und die Lage bei ≤ 160 ° C und niedriger Elektronendosis, sind tiefgefrorenen Gefrierschnitten gefriergetrockneten bei -100 ° C ~ 30 min.
