Für eine Anzahl von Versuchen ist es wichtig, in der Lage, Gewebekultur ex vivo, um die Entwicklung zu verfolgen. Kultur entwickeln Gewebe Zugang zu bestimmten Perioden der Entwicklung und ermöglicht die Manipulation von Genen, die durch die Zugabe von Faktoren, zu dem Kulturmedium, oder beladenen Kügelchen und durch die Verwendung von 1-Transfektion und Elektroporation. Für viele Experimente ist es wichtig, in der Lage, um das Gewebe zu visualisieren, wie es wächst, zum Beispiel, um das Schicksal der Linie markierten Zellen folgen, wie das Gewebe erfährt Morphogenese sein. Dies kann besonders problematisch für Gewebe, die tief innerhalb des Embryos entwickeln, die nicht offensichtlich sind, wenn ein Block von Gewebe aus dem Embryo kultiviert werden. Wir haben eine Live-Scheibe Kulturtechnik ermöglicht es uns, Zahnkeim Entwicklung zu folgen und den Zugang zu den verschiedenen Teilen des Zahnes während der Entwicklung angepasst. Die Technik Kulturen die Zahnkeimscheiben an der Gas-flüssig-Grenzfläche, mit einem modifizierten Verfahren Trowell 3. Diese Scheibe Kulturen wurden direkt nach der Morphogenese des Zahns und damit Linie Verfolgung unterschiedliche Komponenten, wie beispielsweise die Schmelz Knoten und Zahnpapille und Follikel 1,4-7 sehr nützlich. Die Technik ist nicht auf Maus-Embryonen beschränkt und hat erfolgreich die Kultur Live-Scheiben von Schwein und Schlange Zahngewebe 8,9 verwendet. Zusätzlich zu dem Vorteil, dass die Zahnentwicklung sichtbar, weist die Scheibe Verfahren auch den Vorteil, dass die dünnen Scheiben von Gewebe haben Zugang zu Nährstoffen aus dem Medium und Luft aus dem Inkubator erhöht. Dies führt zu einem verbesserten Wachstum der Zahnkeime, die Entwicklung in vivo übereinstimmen, und zeigen INVASIonen von Endothelzellen in die Papille 7. Im Gegensatz Zahnkeime ganz Kiefer Kulturen entwickeln langsamer als die in vivo und die Mitte der Kultur häufig nekrotische in Langzeitkulturen. In Schnittkultur entwickelt der Zahn innerhalb einer Scheibe des Kiefers, und seine Wechselwirkung mit dem umgebenden Knochen zu entwickeln und in anderen Geweben beobachtet werden. In unserem Verfahren wird das Gewebe unmittelbar nach Präparation ohne die Notwendigkeit, in einem Trägermedium 10,11 einzubetten und keine Notwendigkeit für jedes System, das Gewebe an den Hackblock befestigen zerhackt. Das Verfahren ist daher nicht-invasive und schnell, so dass viele Unterkiefer in einer Sitzung geschnitten werden.

Explantatkultur ermöglicht die Manipulation der Entwicklung von Organen zu bestimmten Zeitpunkten und ist damit eine wichtige Methode für die Entwicklungsbiologe. Für viele Organe ist es schwierig, die Entwicklung Gewebe zugreifen Überwachung während der Ex-vivo-Kultur zu ermöglichen. Die Scheibe Kulturverfahren ermöglicht den Zugang zu Gewebe, so daß morphogenetische Bewegungen folgen und spezifischen Zellpopulationen zur Manipulation oder Linienverfolgung richtet sein. In diesem Beitrag beschreiben wir eine Methode der Schnittkultur, die sehr erfolgreich für Kultur der Zahnkeime in einer Reihe von Arten war. Das Verfahren stellt eine hervorragende Anbindung an die Zahnkeime, die mit einer ähnlichen Rate wie in vivo zu beobachten, durch die andere Backe Gewebe umgeben zu entwickeln. Dies ermöglicht Gewebe Wechselwirkungen zwischen dem Zahn und dem umgebenden Gewebe zu überwachen. Obwohl dieses Papier konzentriert sich auf die Zahnkeime, kann das gleiche Protokoll verwendet werden, um die Entwicklung einer Reihe folgenvon anderen Organen, wie Speicheldrüsen, Meckel-Knorpel, Nasendrüsen, Zunge und Ohr.
