Microinjectie is een krachtige techniek voor het leveren van verschillende behandelingen zoals DNA, mRNA, recombinante eiwitten, functieverlies en overexpressie-reagentia, kleurstoffen en combinaties in eieren, embryo&#39;s en cellen van verschillende organismen 1-7. Er moet echter een aantal overwegingen in gedachten worden gehouden bij het ontwerpen van een micro-injectie experiment. Het is uiterst belangrijk om de oplosbaarheid van de geleverde behandeling en het injectievolume...

Efficiënte en reproduceerbare behandeling levering is een van de belangrijkste methodologische uitdagingen voor onderzoekers. Verscheidene methoden zijn vastgesteld om behandelingsoplossingen tijdelijk te leveren in de eieren, embryo&#39;s en cellen. Deze methoden omvatten elektroporatie (gebaseerd op het genereren van tijdelijke poriën in het membraan met behulp van korte elektrische pulsen) 1,2, lipofectie (leverbaar door de fusie van behandeling met liposomen met het membraan) 1, bombardement met microdeeltjes 1 (DNA wordt geprecipiteerd op het micron -sized metalen deeltjes die vervolgens worden gebruikt om de cellen bij hoge snelheid doordringen) en transductie (virus wordt gebruikt als transportmiddel van transgenen). Momenteel, micro-injectie is de enige benadering die het voordeel biedt dat iedere oplossing met 100% rendement met minimale reagentia bezit. Bovendien kan een enkele injectie oplossing bestaat uit een complexe cocktail van behandelingen. Deze techniek is gebruikt om succesvollely microinject de eieren en embryo&#39;s uit tal van soorten, zoals zee-egels 3,4, zebravis 5, 6 muis, kikker 7, en runderen 8, alsmede enkele cellen in het weefsel cultuur 9. Single blastomeren injecties in latere ontwikkelingsstadia zijn ook uitgevoerd 10-12. De huidige werkwijzen voor micro-injecties zijn gebaseerd op de druk-injectiemethode de aanvankelijk beschreven door Hiramoto 10, maar heeft grote vooruitgang geboekt optimalisatie van dit proces. Uitstekende micro-injectie technieken zijn elders 11 beschreven, en hier beschrijven we een van de specifieke methoden die momenteel gebruikt wordt om zee-egel (Strongylocentrotus purpuratus) nieuw bevruchte eieren microinject. Al meer dan een eeuw, hebben zee-egels een waardevol experimenteel model 15,16 geweest. Zee-egels zijn evolutionair nauw verwant aan chordates (waaronder wij) en analyse vanhun genoom onthulde dat ze alle belangrijke gen families als de menselijke 17 bevatten. Ze produceren een groot aantal synchroon ontwikkelen transparante embryo dat gemakkelijk kan worden gemanipuleerd. Het gebruik van zee-egel als modelorganisme, heeft de zee-egel gemeenschap bijgedragen aan ons begrip van de bevruchting proces 18-21, celbiologische processen 22-24, en het gen regulerende netwerken (GRNs) 25-28. Micro-injectie in zee-egel zygotes vereist een aantal stappen. Ten eerste moeten de eieren worden geïmmobiliseerd voor injecties (hieronder beschreven). Micro-injectie gerechten worden bekleed met protamine sulfaat (PS), dat een positief geladen oppervlak waarop de negatief geladen eieren kan hechten 3 ontstaat. De eieren worden dejellied door incubatie in zure zeewater (pH 5.15) gedurende 10 minuten, gevolgd door twee wassingen in natuurlijk zeewater of kunstmatig zeewater (pH 8,0). De dejellied eieren worden zorgvuldig geroeid in een rechte lijnin het midden van PS-gecoate schaal in aanwezigheid van 1 mM 3-aminotriazool (3-AT), die nodig is om de activiteit van ovoperoxidase dat wordt afgescheiden uit de corticale korrels van het ei als gevolg van bevruchting remmen 29. Deze stap is belangrijk om verharding van de bevruchting envelop voorkomen en micro-injectie naald toetreding bewerkstelligen. Als alternatief voor 1 mM 3-AT, kan 10 mM paraminobenzoic zuur (PABA) worden gebruikt. De injectie-oplossing wordt in een micro-injectie naald met behulp van gespecialiseerde microloading pipettip geladen en gemonteerd op een houder bevestigd aan micromanipulator en druk unit (figuur 1). Elke naald kan worden gebruikt om individuele zygoten microinject in meerdere experimenten op verschillende dagen. Micro-injectie kan worden uitgevoerd gedurende 10-15 min tot de zygoten harden. De zygoten worden vervolgens met het zeewater en gekweekte gewassen bij 15 ° C. Wanneer het embryo bereiken uitkomen blastula stadium, geven ze uitbroeden enzym dat onderdelen van de fer verteerttilization envelop 30 en hen in staat stellen om op natuurlijke wijze los te maken van de PS-gecoate schaal. Indien nodig kan de embryo voorzichtig worden losgemaakt van de schaal met een mond pipet of Pasteur pipet zachtjes blazen zeewater op het embryo. De beschreven werkwijze maakt een efficiënte en betrouwbare micro-injectie van 100-400 bevruchte eieren op een schotel, die een high-throughput werkwijze voor downstream analyses.

Micro-injectie in cellen en embryo is een veelgebruikte techniek die wordt gebruikt om een ​​groot aantal biologische processen te bestuderen. Bij deze werkwijze wordt een kleine hoeveelheid behandelingsoplossing in een micro-injectie naald die wordt gebruikt fysiek afzonderlijke injecteren geïmmobiliseerde cellen of embryo geladen.
