In dit protocol kan rustende satellietcellen gemakkelijk worden gezuiverd uit volwassen skeletspier van muizen door collagenase digestie en oppervlakte-antilichaam-gemedieerde MACS scheiding. Deze methode duurt ongeveer 6 uur en geen dure apparatuur zoals een FACS-machine. Daarnaast is deze methode is relatief goedkoop in vergelijking met oppervlakte-antilichaam-gemedieerde FACS scheiding. Een hogere opbrengst van latente satelliet-cellen wordt ook verwacht in vergelijking met FACS voor deze methode omdat FACS laserbeli...

Spier satelliet cellen zijn een kleine populatie van myogene stamcellen zich onder de basale lamina van spiervezels. Ze worden gekenmerkt door de expressie van PAX7, Pax3, c-Met, M-cadherine, CD34, Syndecan-3 en calcitonine 1 - 3. Satelliet cellen bewezen belast spierregeneratie spier stamcellen zijn. Bij volwassen spier-, satelliet-cellen zijn normaal mitotically rust 4-8. Na letsel, zijn satelliet-cellen geactiveerd, initiëren uitdrukking van MyoD, en voer de celcyclus aan hun nageslacht uit te breiden, genoemd myogene voorloper cellen of myoblasts 3. Myoblasten geïsoleerd uit volwassen spier kan gemakkelijk worden uitgebreid ex vivo. De capaciteit van myoblasten tot spiervezels in regenererende spieren envorm buitenbaarmoederlijke spiervezels in nonmuscle weefsels wordt uitgebuit door myoblast transplantatie, een mogelijke therapeutische benadering voor Duchenne spierdystrofie (DMD) 4, urologische dysfunctie 9, en hartfalen 10. Inderdaad, myoblasten met succes getransplanteerd in de spier van zowel mdx (DMD model) muizen en DMD patiënten 11-14. De geïnjecteerde normale myoblasten fuseren met gastheer spiervezels de histologie en functie van de aangetaste spieren verbeteren. Daarom zuivering van een zuivere populatie van latente satelliet cellen van het skelet mu volwassenSCLE is essentieel voor het begrip van de biologie van satelliet-cellen, myoblasts en spierregeneratie, en voor de ontwikkeling van cel-gebaseerde therapieën. De huidige potentiële zuiveringswerkwijzen van latente satelliet cellen vereisen het gebruik van een duur-fluorescentie-geactiveerde celsortering (FACS) machine 1,2,6-8. Bovendien FACS laserbelichting neiging celdood bij scheiding, waarbij lagere opbrengst van latente satelliet-cellen 15 veroorzaakt induceren. Hier presenteren we een nieuwe methode voor de snelle, zuinige en betrouwbare zuivering van latente satelliet cellen uit volwassen muis skeletspieren. Deze werkwijze gebruikt enzymatische dissociatie gevolgd door magnetische-geactiveerde celsortering (MACS). Na isolatie van zuivere rustende satelliet cellen kunnen deze cellen worden gekweekt om grote aantallen myoblasten vinden na een aantal passages. We tonen ook aan dat intramusculaire injectie van deze vers geïsoleerde rustende satelliet cellen of ex vivo geëxpandeerde myoblasten kunnen worden getransplanteerd in cardiotoxine (CTX) geïnduceerde regeneratie muis skeletspieren de bijdrage van donor afgeleide cellen te regenereren spiervezels, alsmede satelliet cel compartimenten voor het onderzoek van zelfvernieuwing-activiteiten.

Spier satelliet-cellen zijn een stamcel populatie vereist voor postnatale skeletspieren ontwikkeling en regeneratie, goed voor 2-5% van sublaminal kernen in spiervezels. Bij volwassen spier-, satelliet-cellen zijn normaal mitotically rust. Na verwonding echter satellietcellen initiëren cellulaire proliferatie myoblasten, hun nakomelingen, de regeneratie van spieren te produceren mediëren. Transplantatie van satelliet-cel afgeleide myoblasten is vaak bestudeerd als mogelijke therapie voor verschillende ziekten waaronder regeneratieve spierdystrofie, hartfalen en urologische stoornissen. Myoblast transplantatie in dystrofe skeletspieren, infarct hart, en disfunctioneren urinewegen heeft aangetoond dat geënt myoblasts kunnen differentiëren in spiervezels in de ontvangende weefsels en tonen gedeeltelijke functionele verbetering bij deze ziekten. Daarom is de ontwikkeling van efficiënte zuiveringswerkwijzen van rustende cellen van satelliet skelet muscle, alsmede de per satelliet cel afkomstige myoblasten culturen en transplantatie werkwijzen voor myoblasten, essentieel voor het begrijpen van de moleculaire mechanismen achter Sat cel zelf-vernieuwing, activatie en differentiatie. Bovendien, de ontwikkeling van cel-gebaseerde therapieën voor spierdystrofie en andere regeneratieve ziekten zijn ook afhankelijk van deze factoren. De huidige potentiële zuiveringswerkwijzen van latente satelliet cellen vereisen het gebruik van dure fluorescentie-geactiveerde celsortering (FACS) machines. Hier presenteren we een nieuwe methode voor de snelle, zuinige en betrouwbare zuivering van latente satelliet cellen uit volwassen muis skeletspier door enzymatische dissociatie gevolgd door magnetische-geactiveerde cel sortering (MACS). Na isolatie van zuivere rustende satelliet cellen kunnen deze cellen worden gekweekt om grote aantallen myoblasten vinden na een aantal passages. Deze vers geïsoleerderustende satelliet cellen of ex vivo geëxpandeerde myoblasten kunnen getransplanteerd worden in cardiotoxine (CTX) geïnduceerde regeneratie muis skeletspieren de bijdrage van donor afgeleide cellen onderzoeken in regenererende spiervezels, alsmede satelliet cel compartimenten voor het onderzoek van zelfvernieuwing activiteiten.

Vers geïsoleerde rustige satelliet cellen vertonen een kleine, ronde vorm (figuur 1G) en een automatische PAX7 als een definitieve marker voor latente satelliet cellen. Meer dan 90% van vers geïsoleerde cellen brengen PAX7 (Figuur 1H en 1I).
