In ons gepresenteerde werk gebruikten we de affiniteitsregistratiebenadering om tether rotavirus DLL's tetheren naar een microfluïdisch platform. Dit maakte in situ beeldvorming van macromoleculaire complexen in een vloeibare micro-omgeving mogelijk. De vangstbenadering is belangrijk met betrekking tot andere microfluïdische beeldvormingstechnieken omdat het biologische exemplaren lokaliseert naar het beeldvormingsvenster om grote diffusieve problemen te ontkrachten die zich voordoen tijdens he...

Een gemeenschappelijk doel van biologen en ingenieurs is om de innerlijke werking van moleculaire machines te begrijpen. Transmissie elektronenmicroscopen (TEM's) zijn ideale instrumenten om deze ingewikkelde details te visualiseren bij bijna-atomaire resolutie1-2. Om het hoogvacuümsysteem van een TEM in stand te houden , zijn biologische monsters doorgaans ingebed in dunne films van glasachtig ijs3, suikers4, zware metaalzouten5, of een combinatie daarvan6. Als gevolg hiervan kunnen afbeeldingen van ingesloten exemplaren slechts beperkte momentopnamen van dynamische processen onthullen.
Vroege pogingen om biologische monsters gehydrateerd in vloeibare omgevingskamers te behouden, werden uitgevoerd door Parsons en collega's met behulp van differentiële pompfasen. Elektronendiffractiepatronen van niet-aangetaste catalasekristallen werden met succes geregistreerd tot een resolutie van 3 Å in een gehydrateerde toestand7-8. Bovendien kunnen fase-gescheiden lipidedomeinen worden onderzocht in gehydrateerde membranen van menselijke erytrocyten9-10. Beweging veroorzaakt door diffusievloeistof en zijn interferentie met de elektronenbundel resulteerde echter in ernstig resolutieverlies en verdere experimenten met biologische monsters werden tot voor kort niet geprobeerd.
Nieuw ontwikkelde microfluïdische monsterhouders zijn geïntroduceerd die halfgeleidermicrochips gebruiken om een microgeschaalde milieukamer te vormen. Deze apparaten kunnen monsters in vloeistof bewaren terwijl ze in eenTEM-kolom 11-12worden geplaatst. Deze technische doorbraak in TEM-beeldvorming heeft onderzoekers in staat gesteld om voor het eerst progressieve gebeurtenissen op moleculair niveau13te bekijken. We verwijzen naar deze nieuwe modaliteit als "in situ moleculaire microscopie" omdat experimenten nu "binnen" deEM-kolom 14-15kunnen worden uitgevoerd. Het algemene doel van deze methode is om biologische assemblages in vloeistof te beelderen om hun dynamische gedrag bij nanometerresolutie te observeren. De reden achter de ontwikkelde techniek is het registreren van real-time waarnemingen en het onderzoeken van nieuwe eigenschappen van biologische machines in oplossing. Deze methodologie breidt het gebruik van TEM's uit voor bredere doeleinden in de cellulaire en moleculaire biologie12-16.
In het huidige videoartikel presenteren we een uitgebreid protocol om een in de handel verkrijgde microfluïdische monsterhouder te assembleren en te gebruiken. Deze gespecialiseerde houders maken gebruik van siliciumnitride microchips geproduceerd met geïntegreerde afstandhouders om een vloeistofkamer te vormen die minuscule hoeveelheden oplossing omsluit. Dunne, transparante ramen zijn geëtst in de microchips voor beeldvormingsdoeleinden12. We demonstreren het juiste gebruik van een microfluïdische houder om simian rotavirus dubbellaagse deeltjes (DLL's) in vloeistof te onderzoeken met behulp van een TEM. Om ervoor te zorgen dat biologische assemblages, zoals DMP's, niet snel over grote afstanden diffuus worden tijdens de beeldvorming, gebruiken we de Affinity Capture-benadering om ze aan het oppervlak van de microfluïdische kamer te vast temaken 16. Deze moleculaire opnamestap heeft een groot voordeel ten opzichte van alternatieve technieken voor het weergeven van biologische monsters in vloeistof, omdat het de verwerving van beelden mogelijk maakt die zullen worden gebruikt voor downstream verwerkingsroutines. Deze opnamestap die wordt gebruikt in combinatie met microfluïdische beeldvorming is uniek voor onze procedures17. Lezers die structurele biologietoepassingen gebruiken met TEM of microfluïdische beeldvormingskamers kunnen het gebruik van affiniteitsopnametechnieken overwegen wanneer dynamische observaties op moleculair niveau het einddoel zijn.

Onderzoekers gebruiken regelmatig Transmission Electron Microscopes (TEMs) om biologische entiteiten te onderzoeken en nieuwe materialen te beoordelen. Hier beschrijven we een extra toepassing voor deze instrumenten- het bekijken van virale assemblages in een vloeibare omgeving. Deze spannende en nieuwe methode voor het visualiseren van biologische structuren maakt gebruik van een recent ontwikkelde op microfluïdische basis gebaseerde monsterhouder. Ons videoartikel laat zien hoe u een microfluïdische houder kunt monteren en gebruiken om vloeibare monsters binnen een TEM in beeld te brengen. In het bijzonder gebruiken we simian rotavirus dubbellaagse deeltjes (DMP's) als ons modelsysteem. We beschrijven ook stappen om het oppervlak van de vloeistofkamer te coaten met affiniteitsbiofilms die DMP's aan het kijkvenster vastklappen. Dit stelt ons in staat om assemblages in beeld te brengen op een manier die geschikt is voor 3D-structuurbepaling. Zo presenteren we een eerste glimp van subvirale deeltjes in een inheemse vloeibare omgeving.

Representatieve afbeeldingen van DMP's in vloeistof met behulp van E-chips die gloeien (figuur 3A) tonen minder DMP's in een bepaald kijkgebied, vermoedelijk als gevolg van diffusie, in vergelijking met DMP's die zijn verrijkt op Affinity Capture-apparaten (Figuur 3B).
