Deze methode werd voor het eerst opgericht in de vroege jaren 1900 1,2 en verwijst naar een techniek waarbij weefselexplantaten werden gekweekt ex vivo in een glazen schaal. Extracellulaire matrix is ​​een integraal onderdeel van het weefsel micro en zijn aanwezigheid is cruciaal voor het handhaven van de fysiologische functies zoals mobiele polarisatie, overleving en proliferatie. Hier presenteren wij een 3-dimensionale weefselkweek methode waarbij primaire beenmerg cells worden gekweekt in extracellulaire matrix geformuleerd om de micro-omgeving van het menselijk bot (RGB-systeem) recapituleren. Ingebed in de extracellulaire matrix worden cellen voorzien van voedingsstoffen door het medium aangevuld met humaan plasma, waardoor een uitgebreid systeem waarbij celoverleving en proliferatie kan worden voortgezet tot 30 dagen terwijl de cellulaire samenstelling van de primaire weefsel. Met behulp van het RGB-systeem hebben we met succes gegroeid primaire beenmergcellen van normale donoren en patiënten met amyloïdose, en diverse hematologische maligniteiten. De RGB-systeem zorgt voor directe, in-matrix real time visualisatie van de cel gedrag en de evaluatie van preklinische werkzaamheid van nieuwe therapieën. Bovendien kunnen cellen worden geïsoleerd uit de rBM vervolgens gebruikt voor in vivo transplantatie, celsortering, stroomcytometrie en nucleïnezuur en eiwitanalyse. Samen genomen, de rBM methode biedt een betrouwbaar systeem voor de groei van de primaire bot Marrow cellen onder fysiologische omstandigheden.

Sinds primaire beenmergcellen niet woekeren onder standaard weefselkweek omstandigheden werd het RGB-systeem gemaakt om het bot micro-omgeving na te bootsen, het verstrekken van een systeem om de cultuur en uitbreiden BM cellen ex vivo.
