De laatste jaren onderzoek erytroblast enucleatie heeft opgedaan toenemende momentum omdat het de stap van in vitro erytropoëse culturen die het meest moeilijk efficiënt reproduceren om succesvol grootschalige productie van rode bloedcellen ex vivo te bereiken. Tot voor kort, de studie van erythroblast enucleatie gebruikt voornamelijk fluorescentie microscopie en flowcytometrie methoden. Fluorescentiemicroscopiemethodes, zij nuttig bij het identificeren deelnemende moleculen vereisen dagen microscopi...

Terminale differentiatie in de erytroïde afstammingslijn bij zoogdieren eindigt met de dramatische proces van enucleatie, waardoor de orthochromatische erytroblast verdrijft het membraan omhulde kern (pyrenocyte) 1, het genereren van een reticulocyten 2. Het exacte mechanisme van deze werkwijze, die ook de snelheidsbeperkende stap van succesvolle, grootschalige productie van rode bloedcellen in vitro, is nog niet volledig opgehelderd. De lokalisatie van de belangrijkste eiwitten en structuren binnen enucleating erythroblasten gebaseerd op het gebruik van fluorescente en elektronenmicroscopie 3-5. Dit moeizaam proces resulteert typisch in de identificatie van een beperkt aantal enucleatie evenementen en niet altijd mogelijk zinvolle statistische analyse. Voortbouwend op een werkwijze erytroblast identificatie eerder beschreven door McGrath et al.. 6, hebben wij een nieuwe benadering voor het identificeren en bestuderen enucleatie gebeurtenissen door Multi-Spectral Ima ontwikkeldGing flowcytometrie (multiparameter snelle celweergave in flow, een methode die fluorescentiemicroscopie met flowcytometrie combineert) 7, dat een voldoende waarnemingen kan leveren uitvoeren van metingen en statistische analyse. Hier beschrijven we eerst twee in vitro erytropoëse kweekmethoden gebruikt om erytroblasten synchroniseren en verhogen de kans op het vastleggen enucleatie bij de evaluatie. Vervolgens beschrijven we in detail de kleuring van erythroblasten na fixatie en permeabilisatie om de lokalisatie van intracellulaire eiwitten of lipide rafts bestudeerd worden enucleatie van multispectrale beeldvorming flowcytometrie. Monsters worden uitgevoerd op een imaging flowcytometer en de verzamelde cellen adequaat gated om orthochromatische erytroblasten 6 identificeren. Orthochromatische erythroblasten worden vervolgens geanalyseerd op basis van hun verhouding, gemeten helderveld imaGing, versus de waarde voor de parameter delta zwaartepunt XY Ter119-DNA, dat wordt gedefinieerd als de afstand tussen de middelpunten van de gebieden, respectievelijk gekleurd voor Ter119 en DNA. De populatie van cellen met een lage aspect ratio en een hoge delta centroïde XY Ter119/DNA is sterk verrijkt in enucleating cellen. Met behulp van wild-type (WT) erytroblasten versus erytroblasten met Mx-Cre-gemedieerde voorwaardelijke schrapping van Rac1 op Rac2 - / - of gecombineerd Rac2 - / -; RAC3 - / - genetische achtergrond en deze nieuwe analyse protocol van multispectrale beeldvorming flowcytometrie, we onlangs aangetoond dat enucleatie lijkt asymmetrische cytokinese en dat de vorming van een ring actomyosine gedeeltelijk gereguleerd door Rac GTPases is belangrijk voor enucleatie progressie 7.

Erytropoëse in zoogdieren wordt afgesloten met het dramatische proces van enucleatie dat resulteert in reticulocyten formatie. Het mechanisme van enucleatie is nog niet volledig opgehelderd. Een veelvoorkomend probleem bij het bestuderen van de lokalisatie van de belangrijkste eiwitten en structuren binnen enucleating erytroblasten microscopisch is de moeilijkheid om een ​​voldoende aantal cellen die enucleatie nemen.
