Flere elegante studier har vist at endotelceller stimulerer neuronal proliferasjon og vice versa ved frigivelse av løselige faktorer, og ved direkte kontakt. Vi har funnet det å vite at i mesteparten av disse studiene 1-3, mens neuronale progenitorer / neurale stamceller isolert fra den embryoniske hjernen, er de cocultured med endotelceller fra den voksne hjernen, andre voksne vev kilder, eller med endotel-cellelinjer. Dette kan delvis skyldes de tekniske problemer forbundet med å isolere og dyrke rene populasjoner av endotelceller fra den embryoniske hjernen. Men, angiogenese, neurogenesis, og neuronal migrasjon er sammenfallende hendelser i størrelsesordener mer robust i den embryonale hjernen enn i den normale voksne hjernen. Den periventricular vaskulære nettverk av embryonic forhjerne (telencephalon) stammer fra et fartøy plassert i basalgangliene primordium og utvikler seg i form av en ordnet gradient fra ventral til dorsal telencephalon ved embryonisk dag 11 (E11) 4,5. Embryonale hjerne endotelceller kan tjene som et viktig verktøy i studiet av angiogenese og nevrovaskulære utvikling og samhandling. I denne fremgangsmåten, blir telencephalon blottet for pial membran oppnådd fra embryoniske dag 15 mus farse, spaltet med kollagenase / dispase, og dispergeres mekanisk i en enkelt cellesuspensjon.
