Celmigratie is een belangrijk aspect voor een studie in kankeronderzoek en het kan ook worden toegepast voor ontwikkeling, immunologische en wondgenezing studies. De celkweek wondsluiting assay en de transwell celmigratie en invasie assays tonen details cel migrerende gedrag en kan worden gebruikt om de moleculaire mechanismen van celmigratie 1,2,10,14 onderzoeken. Beweeglijkheid is een essentieel kenmerk van levende cellen. Celmigratie wordt betrokken bij het ​​ontwerp van het leven, embryonale ontwikkeling, immuunrespons en vele pathologische processen zoals kanker metastase en ontsteking 1-9. Daarom methoden naar cel trekkende gedrag te bestuderen zijn zeer nuttig research tools voor een breed scala van disciplines in de biomedische wetenschappen, biologie, biotechnologie en aanverwante gebieden. De studie van celmigratie in kankeronderzoek is van bijzonder belang als de belangrijkste doodsoorzaak bij kankerpatiënten is gerelateerd aan metastatische progressie. Om voor kanker door het hele lichaam te verspreiden en te verspreiden, moet kankercellen migreren en bedreigen via extracellulaire matrix (ECM), intravasate in de bloedcirculatie, hechten aan een verre plaats, en uiteindelijk extravaseren naar verre brandpunten 1,10-12 vormen. Verschillende biologische methoden kunnen worden toegepast om deze gebeurtenissen in detail te onderzoeken. De celkweek wond-sluiting eend de transwell migratie en invasie assays worden veel gebruikt in de wetenschappelijke gemeenschap 1,10. Werkwijze beweging in beweeglijke cellen kunnen gemakkelijk worden waargenomen met de celcultuur wondsluiting assay. Onder de talrijke manieren om celmigratie te bestuderen, de cel wondsluiting assay is een van de eenvoudigste. Deze werkwijze is nuttig om de migratie vermogen van gehele cellen massa bepalen. Na de analyse van de wondsluiting vele fenotypes kan worden geopenbaard. Het meten van de gesloten afstand in de tijd bij het vergelijken met een controlegroep kan specifieke migratie verandert of een verminderde trekkende fenotype die voorheen onbekend was onthullen. Bovendien kunnen enkele cel lamellipodium vorming, staart intrekken en gerichte beweging aanwijzingen voor wat kan verminderd of versterkt in de cellen van belang 14 geven. De transwell migratie en invasie assays kunnen worden gebruikt om het vermogen van enkelvoudige cellen tot directioneel reageren op de verschillende chemo-aantrekkende of zij chemokines, groeifactoren, lipiden, of nucleotiden 4,5,8,15,16 analyseren. Het kan ook differentiële vermogen tot migratie te beoordelen als gevolg van de over-expressie van een receptor 1,14. Deze testen kunnen ook worden gebruikt om de belangrijkste regulatoren van celmigratie zoals de Rho familie van kleine GTPases 2 te identificeren en karakteriseren. Na deze korte en gemakkelijk acceslijk proeven en de wijze van celmigratie en het vermogen van een cel binnen te dringen in een 3-D-matrix kan ook worden vastgesteld.

in vitro cell migration and invasion assays

Migration is a key property of live cells and critical for normal development, immune response, and disease processes such as cancer metastasis and inflammation. Methods to examine cell migration are very useful and important for a wide range of biomedical research such as cancer biology, immunology, vascular biology, cell biology and developmental biology. Here we use tumor cell migration and invasion as an example and describe two related assays to illustrate the commonly used, easily accessible methods to measure these processes. The first method is the cell culture wound closure assay in which a scratch is generated on a confluent cell monolayer. The speed of wound closure and cell migration can be quantified by taking snapshot pictures with a regular inverted microscope at several time intervals. More detailed cell migratory behavior can be documented using the time-lapse microscopy system. The second method described in this paper is the transwell cell migration and invasion assay that measures the capacity of cell motility and invasiveness toward a chemo-attractant gradient. It is our goal to describe these methods in a highly accessible manner so that the procedures can be successfully performed in research laboratories even just with basic cell biology setup.
