Amoebal coculture og amoebal berikelse er effektive metoder som tillot isolering av mange nye bakterielle og amoebal arter. Resultater oppnådd med disse metodene bekrefter den allestedsnærværende tilstedeværelse av både amøber og Amoeba-motstand bakterier i miljøet, og mest interessant i menneskeskapte vann nettverk som anses for å være styrt av kjemiske behandlinger som klorering og ozonering. Amoebal coculture og amoebal berikelse er viktige verktøy for å isolere og dyrke disse potensielt patogene mikroorga...

Før ankomsten av molekylær diagnose, ble mikroorganismer som er tilstede i miljøet nisjer eller i kliniske prøver ofte påvises ved å dyrke dem på forskjellige selektive medier, hovedsakelig på agar i petriskåler. Fenotyper av bakterielle kolonier og deres metabolsk aktivitet da lov bakteriell klassifisering på artsnivå. Kjøttkraft kan også benyttes for å øke følsomheten for påvisning. Imidlertid har begge teknikkene tillater ikke gjenvinning av bakterier som vokser langsomt eller ikke i det hele tatt i disse media. Dette er grunnen til at molekylære metoder er så mye brukt i dag. Likevel, påvisning av DNA gir ingen anelse om levedyktigheten til bakteriene. Videre contrarily til kulturen, molekylære tilnærminger ikke fører til en belastning som kan bli ytterligere karakterisert. Studerer patogener som vokser dårlig på fast medium eller som trenger celler til å vokse er komplisert. De fleste av disse &quot;vanskelige å vokse&quot; bakterier er kresen intracellulære bakterier, ofte oppdaget og preget etter store utbrudd som det var tilfellet for Legionella pneumophila. Denne bakterien ble preget etter et utbrudd som fant sted under en American Legion konvensjonen. Så mange som 182 personer ble smittet og 29 døde som følge av en alvorlig lungebetennelse 1,2. Det ble senere vist at amøber var de naturlige verter for denne bakterien og at deres tilstedeværelse i hotellet air-condition-systemet og vann nettverk var på opprinnelsen til utbruddet av den såkalte legionella tre. Amøber er til stede over hele verden, og ble isolert fra jord, luft, vann og neseslimhinnen av humane frivillige (gjennomgått i 4). Disse &quot;frittlevende&quot; amøber er generelt dele selvstendig i miljøet, men kan av og til invadere givende vertene fem. Amøber feed på ulike mikroorganismer gjennom fagocytose og påfølgende lysosomal fordøyelsen ved hydrolases seks. Mange fakultative eller obligate intracellulære bakterier er i stand til å motstå fordøyelsen og derved infisere og dividere in amøber som f.eks Legionella, Chlamydia-relaterte bakterier eller mykobakterier (gjennomgått i 7 og 8). Frittlevende amøber sannsynlig representerer en viktig potensielt reservoar for intracellulære bakterier som ennå ikke er oppdaget. Dette førte vår gruppe å gjennomføre i Lausanne to viktigste teknikkene, kalt amoebal coculture og amoebal berikelse, som tillot forskjellige grupper for å isolere flere nye obligat intracellulære mikroorganismer fra ulike miljøprøver 9-15. Siden amøber er profesjonelle fagocytter beite på bakterier, kan en bakterie i stand til å motstå fagocytose og å vokse inni disse protister også kolonisere menneskelige fagocytter og være sykdomsfremkallende mot mennesker. Dette ble delvis påvist for noen Chlamydia-relaterte bakterier, for eksempel Waddlia chondrophila. W. chondrophila kan vokse, ikke bare i amøber, men også i flere celletyper som for eksempel pattedyr epitelceller, makrofager og fiskecellelinjer 16-18. Den amoebal coculture vises også relevant for å påvise intracellulære bakterier i kliniske prøver 19,20, inkludert avføring som er tungt forurenset med ulike bakteriearter 21. Her beskriver vi de viktigste trinnene i amoebal coculture og amoebal berikelse, inkludert (a) behandling av miljømessige eller kliniske prøver, (b) veksten av amøber på axenic media og på en bakteriell plenen av Escherichia coli og (c) valg og karakterisering av intracellulære bakterier.

discovery of new intracellular pathogens by amoebal coculture and amoebal enrichment approaches

Intracellulære patogener som legionella, mykobakterier og Chlamydia-lignende organismer som er vanskelige å isolere, fordi de ofte vokser dårlig eller ikke i det hele tatt på selektive media som vanligvis brukes for å dyrke bakterier. Av denne grunn ble mange av disse patogener oppdaget nylig eller følgende viktige utbrudd. Disse patogener er ofte forbundet med amøber, som tjener som vertscellen, og tillater overlevelse og vekst av bakterier. Vi har tenkt her for å gi en demonstrasjon av to teknikker som tillater isolering og karakterisering av intracellulære patogener til stede i kliniske eller miljøprøver: den amoebal coculture og amoebal berikelse. Amoebal coculture tillater utvinning av intracellulære bakterier ved å inokulere den undersøkte prøven på en amoebal plen som kan være infisert og lysert ved de intracellulære bakterier tilstede i prøven. Amoebal anrikning tillater gjenvinning av amøber tilstede i en klinisk eller miljøprøve. Ther kan føre til oppdagelse av nye amoebal arter, men også av nye intracellulære bakterier som vokser spesielt i disse amøber. Sammen utgjør disse to teknikkene bidra til å oppdage nye intracellulære bakterier i stand til å vokse i amøber. På grunn av deres evne til å infisere amøber og motstå fagocytose, kan disse intracellulære bakterier også unngå fagocytose av makrofager og dermed være patogene for høyere eukaryoter.

Bruke amoebal coculture og amoebal berikelse, ble en hel rekke miljø-og / eller sykdomsfremkallende bakterier oppdaget (Tabell 1). Amoebal coculture ble brukt av vår gruppe og andre til å analysere miljøprøver, vannbehandlingsanlegg og vanndistribusjonssystemer.
