Den store fordelen med CLP er at det tillater forskere å undersøke sepsis av ulike alvorlighetsgrader (dvs. fra lav til middels og høy grad). Sepsis, en systemisk inflammatorisk respons på infeksjon, representerer et ledende dødsårsaken i kritiker syke pasienter en. Intra-abdominale infeksjoner, ofte fører til polymikrobielle sepsis, står for 20% av sepsis tilfeller, som har betydelig dødelighet på opp til 60% 2. Sepsis-assosiert dødelighet primært resultater fra multi-organ dysfunksjon med påfølgende organsvikt 3,4. Ytterligere etterforskning av sykdomsfremkallende mekanismen for denne sykdommen er stort behov for å fremme utvikling av nye og mer effektive behandlingsformer. Den cecal ligation og punktering (CLP)-metoden er en vanlig prosedyre for modellering sepsis in vivo. Som cecum er full av bakterier, dets punktering resultater i polymikrobielle peritonitt, translokasjon av bakterier i blodet (bakteriemi), septisk sjokk, multi-organ dysfunksjon og til slutt døden fem. Det er generelt akseptert at CLP gjenspeiler kliniskvirkeligheten mer nøyaktig enn tidligere teknikker, slik som injeksjon av endotoksin eller rensede bakterier i gnagere, således CLP er ansett gullstandarden (om enn ikke uten begrensninger) 6 for den eksperimentelle induksjon og dermed undersøkelse av patogenesen av sepsis. I denne monografien, beskriver vi protokollene utformet for å vurdere om sykdomsfremkallende mekanismer av sepsis inkluderer autofagi. Autofagi, et evolusjonært konservert cellulære prosessen, forenkler omsetningen av skadede proteiner og organeller som mitokondrier og spiller en viktig rolle i clearance av intracellulære patogener inkludert bakterier 7,8. Under autofagi, er cytosoliske proteiner eller organ trukket inn doble membranbundne vesikler kalt autophagosomes, som senere blir levert til lysosomer for degradering ni. En rekke proteiner har blitt identifisert som pattedyr homologer av autophagy-relaterte gener (ATG),opprinnelig identifisert i gjær, som regulerer prosessen med autophagy. Konverteringen av microtubule assosiert protein-en lett kjede 3B (LC3B) (homologue av Atg8) fra LC3B-I (fri form) til LC3B-II (fosfatidyletanolamin-konjugert form) representerer et stort skritt i autophagosome formasjon 9. Autophagic dysfunksjon er assosiert med aldring og menneskelige sykdommer, inkludert kreft og nevrodegenerative lidelser 10. Videre påvirker autofagi medfødt og adaptiv immunitet som antigen presentasjon, lymfocytter utvikling og cytokin sekresjon av immunceller åtte. Dermed virker det rimelig at autofagi kan også spille en rolle i den systemisk inflammatorisk respons på infeksjon (dvs. i sepsis). Hittil flere metoder har vært beskrevet for å vurdere rollen til autophagy i vevsskade in vivo. Disse inkluderer bruk av grønn fluorescens protein (GFP)-LC3 uttrykker mus og kvantifisering av autophagosomes i vev ved elektronmikroskopi (disse to metoder er beskrevet i monografien). Ytterligere fremgangsmåter omfatter kvantifisering av autophagic proteinekspresjon i vevshomogenater, og analysen av autophagic fluks (som beskrevet andre steder) 11-13. cecal ligation and puncture-induced sepsis as a model to study autophagy in mice

Eksperimentell sepsis kan bli indusert i mus ved hjelp av cecal ligering og punktering (CLP)-metoden, noe som fører til polymikrobielle sepsis. Her blir en protokoll tilgjengelig for å indusere sepsis av varierende alvorlighet hos mus ved hjelp av CLP teknikk. Autofagi er en grunnleggende vev som svar på stress og patogen invasjon. (I) Transgene mus som uttrykker grønt fluorescerende protein (GFP)-LC3 fusjonsprotein blir utsatt for CLP. Studiet av autophagic respons på sepsis er en kritisk component av å forstå de mekanismer som vev reagerer på infeksjon.
