systemic injection of neural stem/progenitor cells in mice with chronic eae

Células tronco / progenitoras neurais (NPCs) são uma fonte de células-tronco promissor para abordagens de transplante, visando a reparação cerebral ou restauração em neurologia regenerativa. Esta directiva surgiu a partir da evidência extensa que a reparação do cérebro é alcançado após o transplante NPC focal ou sistêmica em vários modelos pré-clínicos de doenças neurológicas. Estes dados experimentais identificaram a rota de entrega célula como um dos principais obstáculos de terapias com células estaminais restauradores para doenças cerebrais que requerem avaliação urgente. Enxerto de células-tronco intraparenquimatosa representa uma abordagem lógica para essas patologias caracterizadas por lesões isoladas e acessíveis do cérebro, como lesões da medula espinhal e doença de Parkinson. Infelizmente, este princípio é aplicável mal às condições caracterizadas por uma natureza multifocal, inflamatória e disseminada (no tempo e no espaço), incluindo a esclerose múltipla (MS). Como tal, o cérebro a segmentação por sistentrega NPC émica tornou-se um protocolo invasivo e terapeuticamente eficazes de baixo para entregar células para o cérebro e espinal-medula de roedores e primatas não humanos afectados por danos inflamatória crónica experimental do sistema nervoso central (SNC). Este método alternativo de entrega célula baseia-se na pathotropism NPC, especificamente a sua capacidade inata para (i) detectar o ambiente através de moléculas de adesão de células funcionais e receptores de citocinas inflamatórias e quimiocinas; (Ii) cruzar as barreiras anatômicas vazamento após intravenosa (iv). Ou intracerebroventricular (ICV) de injecção; (Iii) se acumulam no nível do sítio perivascular múltiplo (s) do cérebro e da medula espinhal dano inflamatório; e (iv) exercer notável trófico tecidual e os efeitos de regulação do sistema imunológico em diferentes células alvo hospedeiro in vivo. Aqui vamos descrever os métodos que desenvolvemos para a iv. e &lt;em&gt; entrega icv de NPCs singênicos em camundongos com encefalomielite autoimune experimental (EAE), como modelo de desmielinização inflamatória crônica do SNC, e prevê a entrega de células-tronco sistêmica como uma técnica valiosa para o direcionamento seletivo do cérebro inflamado em neurologia regenerativa.
