dissection of the transversus abdominis muscle for whole-mount neuromuscular junction analysis

上記のプロトコルは、NMJ分析のためのTVA筋肉の分離と染色を指示する。これにより、筋肉のインジュレーションパターンの全体マウント分析と、NMJ形態の高解像度分析が可能です(図2)。この技術は、SMA4,17などの運動ニューロン疾患のマウスモデルにおけるNMJ病理を明らかにするためにうまく応用することができる(図3)。SMAのマウスモデルでは、病理に有意な筋肉内変動があるが、TVA筋肉は一貫して高い影響を受ける。これは、運動エンドプレートの脱用、シナプス前末端および末端発芽における神経フィラメントの蓄積によって証明される(図3)。ここで示した結果は、上記の手法が、マウスモデルにおけるNMJ病理のインナーレーションおよび分析の包括的な概要を提供する強力な方法であることを示す。
