analysis of translation initiation during stress conditions by polysome profiling

שליטה מדויקת של תרגום ה-mRNA היא יסוד להומאוסטזיס תא אוקריוטים, בעיקר בתגובה ללחץ פיסיולוגי ופתולוגיים. שינויים של תכנית זו יכול להוביל לצמיחה של תאים שניזוקו, סימן היכר של התפתחות הסרטן, או למוות של תאים מוקדמים כגון ראה במחלות ניווניות. הרבה ממה שידוע בנוגע לבסיס המולקולרי לבקרת translational כבר מתקבלים מניתוח polysome באמצעות מערכת חלוקה שיפוע צפיפות. טכניקה זו מסתמכת על ultracentrifugation של תמציות cytoplasmic על שיפוע סוכרוז ליניארי. ברגע שהספין הושלם, המערכת מאפשרת חלוקה וכימות של אזורי centrifuged המתאימים לתרגום שונה הריבוזומים אוכלוסיות, ובכך וכתוצאה מכך פרופיל polysome. שינויים בפרופיל polysome מעידים על שינויים או פגמים בייזום תרגום המתרחשים בתגובה לסוגים שונים של מתח. טכניקה זו מאפשרת גם להעריך את התפקיד דואר של חלבונים ספציפיים על ייזום תרגום, וכדי למדוד את פעילות translational של mRNAs הספציפי. כאן אנו מתארים הפרוטוקול שלנו לביצוע פרופילי polysome על מנת להעריך חניכה תרגום של תאים ורקמות אוקריוטים בתנאי גידול נורמלים או מתח.
