Morphogenèse craniofaciale est un processus multi-étapes complexe qui nécessite des interactions coordonnées entre plusieurs types de cellules. La majorité du squelette cranio-facial est dérivé de cellules de la crête neurale, dont beaucoup doivent migrer du tube neural dorsal dans des structures appelées transitoires arcs branchiaux 1. Comme avec de nombreux tissus, la morphogenèse du squelette cranio-facial est plus complexe que ce qui peut être compris par des images statiques d&#39;embryons à des points spécifiques de temps de développement. Moderne, des méthodes pratiques pour la transgenèse et la modification génomique sont progressent rapidement le nombre d&#39;outils disponibles pour les chercheurs de poisson zèbre. Ces outils améliorent les méthodes déjà robustes pour la manipulation génétique et la microscopie. Ces fusions suggèrent fortement que la signalisation BMP dirige la morphogenèse de ces éléments du pharynx. Signalisation PDGF est nécessaire pour le développement cranio-facial, mais a des rôles inconnus en arc branchial morphogenèse. La souris et le poisson zèbre ont mutants PDGFRA profonde clefting midfacial 16-18. Au moins chez le poisson zèbre ce clefting midfacial est due à un échec de la migration des cellules de la crête neurale bon 16. Cellules de la crête neurale continuent d&#39;exprimer PDGFRA après qu&#39;ils ont pénétré dans les arcs branchiaux. En outre, les ligands de PDGF sont exprimés par l&#39;épithélium du visage et dans les arcs branchiaux 16,19,20, ainsi signalisation Pdgf pourraient également jouer un rôle dans la morphogenèse des arcs pharyngiens après la migration. Toutefois, les analyses de la morphogenèse des arcs pharyngiens dans des mutants de PDGFRA n&#39;ont pas été effectuées. analyzing craniofacial morphogenesis in zebrafish using 4d confocal microscopy

Imagerie time-lapse est une technique qui permet l&#39;observation directe du processus de morphogenèse, ou la génération de forme. En raison de leur clarté optique et la susceptibilité à la manipulation génétique, la embryon de poisson zèbre est devenu un organisme modèle populaire avec laquelle effectuer l&#39;analyse time-lapse de la morphogenèse des embryons vivants. L&#39;imagerie confocale d&#39;un embryon de poisson zèbre direct requiert qu&#39;un tissu d&#39;intérêt est constamment marqué avec un marqueur fluorescent, comme un transgène ou d&#39;un colorant injecté. Le processus exige que l&#39;embryon est anesthésié et maintenu en place de manière à ce que le développement sain se déroule normalement. Paramètres pour l&#39;imagerie doivent être définis pour tenir compte de la croissance en trois dimensions et à concilier les exigences de la résolution des cellules individuelles tout en obtenant des clichés de développement. Nos résultats démontrent la possibilité d&#39;effectuer à long terme dans l&#39;imagerie in vivo d&#39;embryons de poisson zèbre de marquées par fluorescence et de détecter des comportements de tissus variés dansla crête neurale crânienne qui causent des anomalies cranio-faciales. Les retards de développement causés par l&#39;anesthésie et le montage sont minimes, et les embryons sont sains et saufs par le processus. Embryons Time-lapse imagées peuvent être retournés pour un milieu liquide et ensuite imagés ou fixés à des points plus tard dans le développement. Avec une abondance croissante des lignes de poisson zèbre transgénique et cartographie sort bien caractérisé et techniques de transplantation, imagerie n&#39;importe quel tissu désiré est possible. En tant que tel, time-lapse imagerie in vivo combine puissamment avec des méthodes génétiques de poisson zèbre, y compris les analyses des embryons mutants et micro-injection.
