Vasektomi är ett relativt enkelt kirurgisk teknik som inte innebär några större svårigheter. Vid preparering med povidon jod och etanol ser till att den sista tvätten (med etanol) avlägsnar povidon jod, eftersom det kan irritera bukhinnan. Tillgången till sädesledaren kan också uppnås genom att pungen eller att utföra en tvärgående snitt i buken 8. Embryotransfer är förmodligen den vanligaste kirurgiska proceduren utfördes i musmodell. Denna teknik är avgörande för att få avkomma från embryon som utsatts för in vitro-manipulation tekniker och utgör därför ett nödvändigt steg för utvecklingen av genetiskt modifierade modeller av pronukleär injektion, lentiviral transduktion eller chimär bildning. Dessutom gör tekniken studiet av utvecklingseffekter av olika förolämpningar som inträffar under preimplantatorisk utveckling. Användningen av konstgjord befruktning 1 eller exponering för onormala halter av olika ämnen eller metaboliter 2 maj påverka embryoutvecklingen resulterar i implantation eller placentation misslyckanden och långsiktiga effekter hos avkomman. En tillförlitlig och reproducerbar embryotransfer teknik är avgörande för att testa eventuella negativa effekter av den experimentella behandlingen om implantation och fosterutveckling på ett konsekvent människaner. Murina preimplantatorisk embryon kan överföras till en mottagare hona antingen in i äggledaren via ampuller på 0,5 dagar efter coitum (DPC) pseudo mottagare (äggledare överföring) 3,4 eller in i livmodern på 2,5 dpc pseudo mottagare (livmoderöverföring) 5,6 beroende på deras utvecklingsstadium. Embryon vid blastocyststadiet, såsom de som användes för att generera chimära möss genom injektion av embryonala eller inducerade pluripotenta stamceller, överförs vanligtvis genom livmoderöverföring. Blastocyster kan också överföras till äggledaren hos en 0,5 dpc mottagaren, men den utgör en mindre fysiologiska test för utvecklingsstörningar störande eftersom embryot genomgår diapaus och har två dagar för att återhämta sig från insult innan implantationen sker. Livmoder byte innebär att punktera livmoderväggen med en smal nål i syfte att alstra en öppning som medger åtkomst av ett embryo manipulation pipetten i livmoderns lumen. ETTlthough denna teknik kan ge goda resultat, (det vill säga andelen embryon som utvecklas till en valp) är överlevnaden på sikt ofta låg och oförutsägbar 7,8. Punkterings av livmoderväggen medför några skadliga biverkningar. För det första är myometrium en mycket vaskulariserad vävnad och dess punktering resulterar ofta i en liten blödning. Blod kan blockera embryotransfer pipett eller invadera livmoderns lumen orsakar embryonal död och / eller implantation misslyckande. Detta är särskilt relevant när embryon utan zona överförs, eftersom blodkroppar och skräp kan fästa till blastomererna. För det andra gör öppningen utförs inte tätar efter embryon har överförts, så att de kan flyta tillbaka genom öppningen och utvisas till bukhålan då en för stor volym har introducera in i livmodern. För att få en mottagare, är högst 2 honor av 8 veckor till 6 månaders ålder placeras med en vasektomiserade hane på eftermiddagen. Följande morgon, är honor kontrolleras med avseende på förekomst av en vaginal samlag plugg, en klump koagulerat protein från den manliga sädesvätskan. Som parning inträffar vanligen under midnatt, är dagen för vaginal plugg upptäckt anses vara 0,5 DPC. Även vasektomiserade hanar kan köpas från vissa leverantörer, är relativt lätt det kirurgiska ingreppet som beskrivs här och inte kräver några ytterligare instrument än vad som krävs för embryoöverföring.

Överföringen av preimplantatorisk embryon till en surrogat hona är ett obligatoriskt steg för produktion av genetiskt modifierade möss eller att studera effekterna av epigenetiska förändringar har sitt ursprung under preimplantatorisk utveckling på senare fosterutveckling och vuxenhälsa. Användningen av en effektiv och konsekvent embryotransfer teknik är avgörande för att stärka den generation av genetiskt modifierade djur och att fastställa effekten av olika behandlingar på implantation priser och överlevnad på sikt.
