Peptid tertiära amider (pesetas) är en superfamilj av peptidomimetiska oligomerer. Förutom de väl studerade peptider, peptoider och N-metylerade peptider, förblir en stor del av föreningarna inom denna familj understudied, majorly grund av brist på syntetisk metod för att komma allmänna N-alkylerade peptider. Peptidhärmande är föreningar, som efterliknar strukturen av naturliga peptider. De är utformade för att bevara bioaktiviteten samtidigt övervinna några av de problem som är förknippade med naturliga peptider, inklusive cell permeabilitet och stabilitet mot proteolys 1-3. På grund av den oligomera karaktären hos dessa föreningar kan stora syntetiska bibliotek lätt nås genom monomera eller sub-monomera syntesvägar 4-7. En av de mest studerade klasserna av peptidomimetiska är peptoids. Peptoider är oligomerer av N-alkylerade glyciner som kan syntetiseras enkelt med hjälp av en sub-monomer strategi 8, 9. Många användbara proteinligander har framgångsrikt identifierat från screening stora syntetiska peptoid bibliotek mot protein mål 1, 10-14. Ändå &quot;träffar&quot; som identifierats från peptoid bibliotek arkivera sällan mycket hög affinitet mot proteinmål 1,10-14,22. En major skillnaden mellan peptoider och naturliga peptider är att de flesta av peptoider allmänhet saknar förmågan att bilda sekundärstruktur på grund av avsaknaden av kirala centra och konformationsbegränsningar. Peptid tertiära amider (pesetas) är en superfamilj av peptidhärmande molekyler som inkluderar men inte är begränsade till peptider (R 2 = H), peptoider (R 1 = H) och N-metylerade peptider (R 1 ≠ H, R2 = Me) . (Se figur 1) Vårt syntesväg sysselsätter naturligt förekommande aminosyror som källa för chiralitet och sidokedjorna på45;-kol, och kommersiellt tillgängliga primära aminerna för att ge N-substitutioner. Därför kan en större kemisk utrymme än för enkla peptider, peptoids eller N-metylerade peptider utforskas. Cirkulär dikroism spektra har visat att PTA molekyler är mycket strukturerad i lösning. Karakterisering av en av de PTA-proteinkomplex visar tydligt att de konformationsbegränsningar för tereftalsyra krävs för bindning. Nyligen har vi också upptäckt att vissa av PTA molekyler har förbättrat cellerpermeabilitet än sina peptoid och peptid motsvarigheter. Vi tror att dessa PTA bibliotek kan vara en bra källa till hög-affinitetsligander för protein mål. split-and-pool synthesis and characterization of peptide tertiary amide library

Peptidhärmare är bra källor till protein-ligander. Den oligomera karaktären av dessa föreningar gör att vi kan få tillgång till stora syntetiska bibliotek på fast fas med hjälp av kombinatorisk kemi. En av de mest studerade klasser av peptidomimetiska är peptoids. Peptoider är lätta att syntetisera och har visat sig vara proteolys beständigt och cell-permeabel. Under det senaste decenniet har många användbara proteinligander identifierats genom screening av peptoid bibliotek. Men de flesta av de ligander som identifierats från peptoid bibliotek visar inte hög affinitet, med få undantag. Detta kan bero delvis på bristen på kirala centra och konforma begränsningar i peptoid molekyler. Nyligen beskrev vi en ny syntetisk väg att komma peptid tertiära amider (pesetas). Trafikhuvudmän är en superfamilj av peptidomimetika som inkluderar men är inte begränsade till peptider, peptoider och N-metylerade peptider. Med sidokedjor på både α-kol och huvudkedjan kväveatomer,konforma av dessa molekyler är kraftigt begränsade av steriska hinder och allyliska 1,3 stammen. (Figur 1) Vår studie tyder på att dessa PTA molekyler mycket strukturerad i lösning och kan användas för att identifiera proteinligander. Vi tror att dessa molekyler kan vara en framtida källa av hög affinitet proteinligander. Här beskriver vi den syntetiska metoden som kombinerar kraften i både split-och-pool och strategier sub-monomera att syntetisera ett prov en pärla en förening (OBOC) bibliotek med trafikhuvudmän.

Här visar vi tre representativa MALDI spektrum från en PTA trimer med länkaren. Såsom visas i figur 6A, när klyvas under rumstemperatur med användning av 50% TFA / DCM-lösning, signifikant nedbrytning observerades. I figur 6A, topp 593 och 484 motsvarar länkaren och PTA trimeren respektive, visar att hela molekylen framgångsrikt syntetiseras på pärla men bryts ner vid klyvning.
