De stappen in het hierboven beschreven protocol een leidraad hoe voor te bereiden hippocampus acute gesneden uit jonge of volwassen ratten, opwekken en opnemen LTP, snel ontleden gestimuleerde gebied van slice, en voor te bereiden nucleaire verrijkte fractie voor activiteit afhankelijk eiwit dynamiek te bestuderen. Deze benadering is afgeleid van een combinatie van verschillende methoden onafhankelijk van elkaar gebruikt. Synaptic N-methyl-D-aspartaat-receptoren (NMDARs) spelen een cruciale rol in synaptische plasticiteit en celoverleving signaleren dat activatie van extrasynaptic NMDARs kan leiden neurodegeneratie en celsterfte. Deze veranderingen hangen af van streng gecontroleerde / gereglementeerde activiteit afhankelijke genexpressie en dus vereisen constante communicatie tussen geactiveerde synapsen of dendrieten en de kern 7. De MAP kinases ERK1 / 2 zijn effectoren van synaptische NMDARs signalering en zijn betrokken bij NMDAR-activatie-geïnduceerde genexpressie, dat signalering via extrasynaptic NMDAR geen of een remmend effect op de ERK1 / 2 activiteit 8,11. Er zijn een aantal eiwitten die zijn aangetoond shuttle tussen distale dendrieten en de nucleus. Veel van deze eiwitten een nucleair lokalisatiesignaal en actief aan de kern 6,9 getransporteerd langs microtubuli in een dynein en importin afhankelijke wijze. Interestingly, sommige boodschappers alleen worden doorgevoerd naar de kern in reactie op specifieke synaptische stimuli. Bijvoorbeeld wordt retrograde transport van cyclisch AMP respons element bindend eiwit 2 (CREB2) geïnduceerd door chemische LTD maar niet LTP 12. Gelokaliseerde NMDAR synaptische stimulatie drijft-CREB gereguleerde transcriptie coactivator (CRTC1) in de kern, een translocatie proces, dat betrokken is bij de lange termijn hippocampale plasticiteit 4. Recent werd aangetoond dat het eiwit boodschapper Jacob naar de kern nadat beide, synaptische en extrasynaptic NMDAR activatie en regelt CREB afhankelijke gentranscriptie 5. De synaptische of extrasynaptic oorsprong van het signaal gecodeerd in een posttranslationele modificatie van Jacob. Synaptische activiteit induceert ERK1 / 2 afhankelijke fosforylatie van Jacob in een cruciale serine op positie 180 (pJacobS 180) dat een voorwaarde voor de daaropvolgende translocatie naar de kern van primaire hippocampale kweek. Bovendien in CA1 neuronen van acute hippocampus plakjes pJacobS 180 naar de kern na Schaffer onderpand LTP maar niet LTD 1,10. pS180 Jacob leidt tot een verhoogde expressie van plasticiteit gerelateerde genen en deze genexpressie terugvoert synaptische functie. In scherp contrast, Jakob die naar de kern na extrasynaptic NMDARs activering niet is gefosforyleerd op Ser180 en kan worden geassocieerd met verschillende eiwit complex in de kern veroorzaken &quot;CREB uitgeschakeld en een retractie van synaptische contacten 10. De meeste gepubliceerde studies over de nucleaire import van synapto-nucleaire eiwit boodschapper zijn gedaan in gedissocieerde neuronale primaire culturen. Daarom zou het interessant zijn om te zien of dergelijke bevindingen kunnen worden gereproduceerd in fysiologisch relevante omstandigheden met behulp van de hippocampus plakjes waar neuronale connectiviteit en functie zijn veel beter bewaard gebleven. Hier presenteren we een geoptimaliseerd protocol voor de beoordeling van LTP-deaanhangende nucleaire translocatie van eiwitten boodschappers door immunoblotting. Deze methode is ook geschikt voor het analyseren van activiteit afhankelijke fosforylering van proteïnen in een ruw nucleaire fractie. Specifiek, het huidige protocol omvat de voorbereiding van acute CA1 hippocampus plakjes, inductie, en de opname van LTP. Vervolgens wordt CA1 regio microscopisch ontleed om de gestimuleerde gebied isoleren. Wij gecombineerd en bewerkt het protocol voor nucleaire isolatie door CellLytic nucleaire Extraction Kit met veranderingen die door Zhao en collega&#39;s 17. De geoptimaliseerde procedure omvat de lysis van ontleed CA1 regio hypotone buffer waardoor cel zwelling en afgifte van kernen. Cell lysis en kernen morfologie kan worden bepaald door microscopisch onderzoek. Nuclear verrijking bereikt door een korte centrifugatie stap. Immunoblotting analyse met antilichamen tegen NeuN en NSE2 specifieke merkers van nucleaire of cytosolische fracties, geeft aan dat deze benadering kan worden gebruikt als een snelleen reproduceerbare protocol deze subcellulaire fracties te isoleren en zeer labiele posttranslationele modificaties zoals eiwitfosforylatie bestuderen. Bovendien is deze werkwijze voordelig voor kleine weefselmonsters afkomstig van ontleed CA1 regio van hippocampale plakjes en kan worden gebruikt in combinatie met immunohistochemie van hippocampale plakjes.

isolation of ca1 nuclear enriched fractions from hippocampal slices to study activity-dependent nuclear import of synapto-nuclear messenger proteins

Bestuderen activiteit afhankelijke eiwitexpressie subcellulaire translocatie of fosforylering essentieel om de onderliggende cellulaire mechanismen van synaptische plasticiteit begrijpen. Lange termijn potentiëring (LTP) en lange termijn depressie (LTD) geïnduceerde acute hippocampale plakken worden algemeen aanvaard als cellulaire modellen van leren en geheugen. Er zijn tal van studies die live cell imaging of immunohistochemie benaderingen gebruiken om de activiteit afhankelijk eiwit dynamiek te visualiseren. Echter deze werkwijzen afhankelijk van de geschiktheid van antilichamen voor immunocytochemie of overexpressie van fluorescentie-gemerkte eiwitten in enkele neuronen. Immunoblotting van eiwitten is een alternatieve methode voor het verstrekken van onafhankelijke bevestiging van de bevindingen. De eerste bepalende factor voor de bereiding van subcellulaire fracties individueel tetanized hippocampale plakken is de lage hoeveelheid materiaal. Ten tweede, de behandeling procedure is cruciaal omdat zelfs zeer korte en kleine manipulaties living plakjes zou activering van bepaalde signaleringscascades induceren. Hier beschrijven we een optimale workflow om voldoende hoeveelheid nucleaire verrijkte fractie van voldoende zuiverheid te verkrijgen van de CA1 regio acute hippocampale plakjes van rattenhersenen. Als een representatief voorbeeld wordt aangetoond dat de ERK1 / 2 gefosforyleerde vorm van het synapto-kerneiwit messenger Jacob actief naar de kern na inductie van LTP en kan worden gedetecteerd in nucleaire verrijkte fractie uit CA1 neuronen.

We hebben eerder aangetoond dat de synapto-kerneiwit messenger Jacob accumuleert in de kern na de inductie van LTP maar niet LTD 1. Bovendien translocatie van Jacob na synaptische stimulatie vereist activatie van MAPK ERK1 / 2 en fosforylering van Jacob op Ser180 (figuur 1).
