In Pflanzen spielen wechselnden mechanischen Eigenschaften eine wichtige Rolle bei der Leitung Wachstum und Morphogenese. Bis heute hat es große Fortschritte in der Entschlüsselung der genetischen und chemischen Netzwerke, die das Pflanzenwachstum zu kontrollieren, aber unser Wissen, wie diese Netzwerke beitragen, und sind durch Veränderungen der mechanischen Eigenschaften betroffen ist rudimentär. Diese Methode sollte es uns ermöglichen, diese Lücke zu füllen, und so sollte es ein starkes Interesse an Wissenscha...

Wachstum der Pflanzen wird durch die koordinierte Ausdehnung der starren Zellwänden, die jede einzelne Zelle des Organismus umgibt erreicht. Zunehmend Hinweise darauf, zeigt, dass es durch die Modifikation der Zellwand der Chemie, dass Pflanzen lokal diese Expansion zu steuern. Die Expansion wird angenommen, dass in erster Linie durch Druck auf den Zellwänden von hohen Turgor der Zelle verursacht angetrieben zu werden; Dieser Stamm Reaktion auf Turgor wird durch die mechanischen Eigenschaften der Zellwände 1 geregelt. Wenig ist von dieser mechanischen Eigenschaften bekannt und wie sie während der Entwicklung ändern. Weiterhin ist wenig wie diese mechanischen Eigenschaften gesteuert werden, und ob erteilen tragen zur Zellwandchemie in einer Weise, die offensichtlich in einer Gewebe koordiniert verändern bekannt. Wenn wir die Verbindung zwischen chemischen und mechanischen Veränderungen in der Pflanzenzellwände während der Entwicklung, und schließlich, wie diese mikroskopischen Wechselwirkungen regieren ein Werk verstehen&#39;S makroskopischen Wachstums, eine Methode, die die mechanischen Eigenschaften der Zellwände bei der Entwicklung von Organen an der Zell-oder Gewebe Maßstab überwachen erforderlich. Die Rasterkraftmikroskopie (AFM)-Methode hier beschrieben, die auf Mikrometer-oder Nanometergewebekompressionen oder Vertiefungen basiert, wurde entwickelt, um genau die mechanischen Eigenschaften der Zellwände in der Entwicklung gleichzeitig Organe auf subzellulärer Auflösungen und über ganze Regionen des Gewebes zu messen. Andere Methoden entweder eine Auflösung, die zu niedrig oder zu hoch ist: das Extensometer ist nur in der Lage, die durchschnittlichen mechanischen Eigenschaften eines ganzen Gewebes an der Millimeter-Skala 2-4, einer Skala, die beispielsweise zu groß, um in frühen Ereignisse zu messen, ist zu messen Organogenese; die Mikroindenter können Messungen an subzellulärer Auflösung im Nanometerbereich, aber es wird in den Messzellen isoliert und nicht Gruppen von Zellen oder Organen 5-7 beschränkt. Mit der AFM, die erforderlichd Gewebe, Zell und subzellulärer Auflösungen 8-10 erreicht werden. Kürzlich mehrere Protokolle wurden speziell entwickelt, um Mechanismen der Gewebe Pflanzen, die auch verwendet werden könnte, 11, 12 zu messen. Wir werden hier präsentiert, wie die Elastizität des Gewebes durch Messung der scheinbaren Elastizitätsmodul 13 zu beurteilen. Der Elastizitätsmodul wird häufig verwendet, um die Steifigkeit eines Materials zu beschreiben. Während kleine Verformung der erforderlich ist, um ein Material zu verformen Kraft ist proportional zur Fläche des Eindrucks. Der E-Modul ist dieser Koeffizient. Im Fall einer kontinuierlichen homogenen Material die gleichen Koeffizienten unabhängig von der Einrückung Typ (Größe und Form) gemessen werden, jedoch wird mit der Geschwindigkeit der Messung verändern. Im Falle der komplexen Struktur von Pflanzen Gewebe haben wir so weit, dass die Kraft proportional zu der Deformation, die die Bestimmung der beobachtetenein Koeffizient der Verhältnismäßigkeit, die wir nennen &quot;scheinbare Elastizitätsmodul&quot;. Im Gegensatz von kontinuierlichen Medien in den Pflanzen ist diese scheinbare Elastizitätsmodul empfindlich auf die Größe der Vertiefung. Es muss nicht auf die jungen Moduli eines reinen Zellwand entsprechen. Es beschreibt die beste Elastizität des Gerüsts der Zellwand des Gewebes.

afm-based mapping of the elastic properties of cell walls: at tissue, cellular, and subcellular resolutions

Wir beschreiben eine kürzlich entwickelte Methode, um die mechanischen Eigenschaften der Oberflächen von Pflanzengeweben für ein JPK AFM-Messung mit Hilfe der Rasterkraftmikroskopie (AFM) Mikro / Nano-Vertiefungen. Insbesondere wird in diesem Protokoll wir das scheinbare Elastizitätsmodul der Zellwände zu messen subzellulären Auflösungen in den Regionen von bis zu 100 &amp;mgr; m × 100 &amp;mgr; m in Blütenmeristemen, Hypokotylen und Wurzeln. Dies erfordert eine sorgfältige Vorbereitung der Probe, die richtige Auswahl der Mikro-Eindringkörper und Eindringtiefen. Um die Zellwand nur Eigenschaften werden Messungen in hochkonzentrierten Lösungen von Mannitol, um die Zellen plasmolysieren beseitigen und damit den Beitrag der Zelle Turgor geführt ausmachen. Im Gegensatz zu anderen vorhandenen Techniken, durch die Verwendung unterschiedlicher Eindringkörpern und Eindringtiefen ermöglicht dieses Verfahren die gleichzeitige Multiskalen-Messungen <em&gt; Dh bei subzellulärer Auflösungen und über Hunderte von Zellen, die ein Gewebe. Dies bedeutet, dass es nun möglich ist, räumlich-zeitlich charakterisieren die Veränderungen in den mechanischen Eigenschaften der Zellwände während der Entwicklung zu nehmen, so dass diese Änderungen mit Wachstums-und Differenzierungs korreliert werden. Damit ist ein wichtiger Schritt, um zu verstehen, wie koordinierte mikroskopischen Zellveränderungen bewirken makroskopischen morphogenetischen Ereignisse. Es bleiben jedoch mehrere Einschränkungen: Das Verfahren kann nur für relativ kleine Proben (etwa 100 um Durchmesser) und nur an äußeren Gewebe verwendet werden; die Methode ist empfindlich, um Gewebe Topographie; sie misst nur bestimmte Aspekte des Gewebes komplexen mechanischen Eigenschaften. AFM-based Mapping of the Elastic Properties of Cell Walls: at Tissue, Cellular, and Subcellular Resolutions

We describe a method to map mechanical properties of plant tissues using an atomic force microscope (AFM). We focus on how to record mechanical changes that take place in cell walls during plant development at wide-field mesoscale, enabling these changes to be correlated with growth and morphogenesis.
