Crescita nelle piante è ottenuta dall&#39;espansione coordinata delle pareti cellulari rigide che circondano ogni cellula dell&#39;organismo. Accumulando prove indicano che è attraverso la modifica della chimica parete cellulare che le piante controllano localmente questa espansione. L&#39;espansione è pensato per essere determinata principalmente da sforzo sulle pareti cellulari, causate da alta pressione turgore della cellula; questa risposta ceppo di turgore è regolato dalle proprietà meccaniche delle pareti cellulari 1. Si sa poco di queste proprietà meccaniche e come cambiano durante lo sviluppo. Inoltre si sa poco di come queste proprietà meccaniche sono controllate e se le risposte contribuiscono ad alterare la chimica della parete cellulare in un modo che è apparentemente coordinata tra un tessuto. Se vogliamo comprendere la connessione tra cambiamenti chimici e meccanici nelle pareti cellulari delle piante durante lo sviluppo, e in ultima analisi come queste interazioni microscopiche governare una piantaÈ necessario s &#39;crescita macroscopica, un metodo che può monitorare le proprietà meccaniche di pareti cellulari nello sviluppo organi a scala cellulare o tessutale. Il metodo di microscopia a forza atomica (AFM) qui descritto, che si basa su micrometriche o nanometriche compressioni tessuto o rientranze, è stato sviluppato proprio per misurare le proprietà meccaniche delle pareti cellulari nello sviluppo organi contemporaneamente a risoluzioni subcellulari e attraverso intere regioni di tessuto. Altri metodi hanno o una risoluzione che è troppo basso o troppo alto: l&#39;estensimetro è solo in grado di misurare le proprietà medie meccaniche di un intero tessuto alla scala millimetrica 2-4, una scala che è ad esempio troppo grande per misurare eventi precoci organogenesi; il microindenter può prendere misure a risoluzione subcellulare su scala nanometrica, ma si è limitato a misurare cellule isolate e non gruppi di cellule o organi 5-7. Con l&#39;AFM, l&#39;richiedonod tessuto, cellulare, e risoluzioni subcellulari possono essere raggiunti 8-10. Recentemente diversi protocolli sono stati sviluppati specificamente per misurare meccanica di piante tessuto che potrebbero anche essere utilizzati 11, 12. Nel seguito verrà illustrato come valutare l&#39;elasticità del tessuto attraverso la misurazione della apparente modulo di Young 13. Il modulo di Young è comunemente usato per descrivere la rigidità di un materiale. Durante piccola deformazione la forza necessaria per deformare un materiale è proporzionale all&#39;area di rientro. Il modulo di Young è tale coefficiente. Nel caso di un materiale omogeneo continua lo stesso coefficiente sarà misurato indipendentemente dal tipo rientranza (dimensioni e forma) ma cambia con la velocità della misurazione. Nel caso della struttura complessa di piante tessuto, abbiamo osservato finora che la forza è proporzionale alla deformazione permettendo la determinazioneun coefficiente di proporzionalità che chiamiamo &quot;giovani modulo apparente&quot;. In contrasto da continue medias nelle piante, questa apparente modulo di Young è sensibile alla dimensione della rientranza. Non corrisponde alla giovane moduli di una parete cellulare puro. Esso descrive meglio l&#39;elasticità del ponteggio della parete cellulare del tessuto.

afm-based mapping of the elastic properties of cell walls: at tissue, cellular, and subcellular resolutions

Descriviamo un metodo recentemente sviluppato per misurare le proprietà meccaniche delle superfici dei tessuti vegetali utilizzando microscopia a forza atomica (AFM) micro / nano-rientranze per un JPK AFM. In particolare, in questo protocollo misuriamo modulo apparente di Young delle pareti cellulari con risoluzione subcellulare in tutte le regioni fino a 100 micron x 100 micron di meristemi floreali, ipocotili e radici. Questo richiede un&#39;attenta preparazione del campione, la corretta selezione di micro-penetratori e profondità di rientro. Per spiegare le proprietà della parete cellulare solo, le misurazioni vengono eseguite in soluzioni altamente concentrate di mannitolo per plasmolyze le cellule e rimuovere il contributo del turgore cellulare circoscritta. A differenza di altre tecniche esistenti, utilizzando diversi penetratori e profondità di rientro, questo metodo consente misurazioni multiscala simultanea, <em&gt; Cioè a risoluzioni subcellulare e su centinaia di cellule che compongono un tessuto. Ciò significa che è ora possibile caratterizzare spazialmente-temporalmente i cambiamenti che avvengono nelle proprietà meccaniche dei pareti cellulari durante lo sviluppo, consentendo a questi cambiamenti essere correlato con la crescita e la differenziazione. Questo rappresenta un passo fondamentale per capire come i cambiamenti cellulari microscopiche coordinati portare a eventi morfogenetici macroscopici. Tuttavia, diverse limitazioni restano: il metodo può essere usato solo abbastanza piccoli campioni (circa 100 micron di diametro) e solo su tessuti esterni; il metodo è sensibile alla topografia tessuto; misura solo alcuni aspetti del complesso proprietà meccaniche del tessuto. La tecnica è stata sviluppata rapidamente ed è probabile che la maggior parte di queste limitazioni saranno risolti nel prossimo futuro.

AFM-based Mapping of the Elastic Properties of Cell Walls: at Tissue, Cellular, and Subcellular Resolutions

We describe a method to map mechanical properties of plant tissues using an atomic force microscope (AFM). We focus on how to record mechanical changes that take place in cell walls during plant development at wide-field mesoscale, enabling these changes to be correlated with growth and morphogenesis.
