afm-based mapping of the elastic properties of cell walls: at tissue, cellular, and subcellular resolutions

Vi beskriver en nyligen utvecklad metod för att mäta mekaniska egenskaper hos ytorna av växtvävnader med hjälp av atomkraftsmikroskopi (AFM) mikro / nanointryckningar, för en JPK AFM. Närmare bestämt i detta protokoll mäter vi den skenbara elasticitetsmodulen av cellväggarna på subcellulära upplösningar i olika regioner på upp till 100 ^ m x 100 | im i floral meristem, hypokotyler och rötter.
