Het doel van deze huidige protocol is licht te werpen op de (soms) ingewikkelde procedures die to de synthese en biochemische karakterisatie van deze geactiveerde bouwstenen (Figuur 1B). Speciale aandacht zal worden gegeven op alle synthetische details die vaak moeilijk te vinden zijn of afwezig zijn in de experimentele afdelingen maar nog cruciaal voor de succesvolle afronding van de synthetische weg die leidt tot de isolatie van zuiver (d) NTP&#39;s (figuur 1).

De traditionele strategie voor de introductie van chemische functionaliteiten is het gebruik van vaste-fase synthese door het toevoegen van geschikte gemodificeerde fosforamidiet precursors naar het ontluikende keten. Echter, de gebruikte tijdens de synthese en de beperking tot vrij korte sequenties omstandigheden belemmeren de toepasbaarheid van deze methodiek. Anderzijds worden gemodificeerd nucleoside trifosfaten geactiveerd bouwstenen die zijn gebruikt voor de licht invoering van talrijke functionele groepen in nucleïnezuren, een strategie die de weg vrijmaakt voor het gebruik van gemodificeerde nucleïnezuren in een breed palet van praktische toepassingen zoals functionele tagging en genereren van ribozymen en DNAzymen. Een van de grote uitdagingen bevindt in de complexiteit van de methode leidt tot de isolatie en karakterisering van deze nucleoside analogen. In deze video artikel presenteren we een gedetailleerd protocol voor de synthese van These gemodificeerde analogen met fosfor (III)-gebaseerde reagentia. Bovendien is de procedure voor de biochemische karakterisatie openbaar gemaakt, met bijzondere aandacht voor primerverlengingsreacties en TdT tailing polymerisatie. Deze gedetailleerde protocol zal van nut zijn voor het vervaardigen van gemodificeerde dNTPs en het gebruik daarvan in de chemische biologie.

Gemodificeerde nucleosidetrifosfaten zijn verleidelijke synthetische doelen, omdat zij zorgen voor de gemakkelijke invoering van een breed scala van functionele groepen in nucleïnezuren 41. Echter, de isolatie en karakterisering van deze geactiveerde bouwstenen vaak lastig blijkt te zijn. Bijgevolg is de hierin getoonde resultaten worden verondersteld om een helpende hand te bieden aan de verschillende stappen binnen de genoemde synthetische en biochemische procedures (Figuur 1B) te volgen. ...
