De bevolking van microRNA vormt ongeveer 0,4-0,5% van het totaal RNA in het serum gevonden. Verder is er ook een hoog eiwitgehalte in humaan serum. In de afgelopen jaren is er een groeiende druk om nieuwe biomarkers te ontdekken voor de vroege opsporing van ziekten bij de mens. Veel aandacht is gericht op het gebruik van kleine RNA&#39;s zoals microRNA 2 (miRNAs of miRs) als potentiële markers. Deze kleine RNAs worden aangetroffen in lichaamsvloeistoffen zoals serum en studies hebben aangetoond ze bestand tegen afbraak en stabiel zijn over een bereik van variërende omgevingsomstandigheden 3. Gezien deze kenmerken, serum of circuleren miRNAs zijn de ideale biomarker 4, 5. Momenteel zijn er twee belangrijke benaderingen voor het isoleren van kleine RNA uit biologische vloeistoffen. De eerste benadering maakt gebruik van kolommen gebaseerde technologie te binden en elueren de kleine RNAs 6, terwijl de tweede benadering gebruikt de langdurige protocol met fenol en guanidine thiocyanaat reagentia 7. We hebben een eenvoudig, effectief, kolomvrije protocol om kleine RNA te isoleren uit humaan serum ontwikkeld. Het geïsoleerde RNA is onmiddellijklijk bruikbaar in afgeleide toepassingen, met inbegrip van DNA-oligonucleotide arrays en RNA sequencing. Dit protocol is ontwikkeld omdat we werden geconfronteerd met een aantal problemen bij het gebruik van fenol gebaseerde methoden voor het isoleren van RNA uit serum. De traditionele benadering Chomczynski wordt vaak gebruikt in de meeste laboratoria met verschillende reagentia verkrijgbaar bij de meeste commerciële verkopers. Maar gezien de grootschalige toepassing ervan, strenge richtlijnen zijn niet ontwikkeld om consequent produceren hoogwaardige RNA van lichaamsvloeistoffen, met name bloed of serum. Veel voorkomende problemen in verband met het isoleren van RNA uit het serum onder lage opbrengsten RNA en besmetting met reagentia gebruikt tijdens de isolatie, met name fenol. Onze aanpak elimineert deze fenol verontreinigingen van hoge kwaliteit RNA voor downstream-analyse zoals kwantitatieve PCR (qPCR) en RNA sequencing. We hebben verder getest dit RNA op miRNA arrays.

De analyse van RNA en de expressie ervan is een gemeenschappelijk kenmerk in veel laboratoria. Van belang is het ontstaan ​​van kleine RNAs, zoals microRNAs, die worden aangetroffen in zoogdiercellen. Deze kleine RNA&#39;s zijn krachtige gen regelgevers beheersen van vitale routes zoals groei, ontwikkeling en dood en veel belangstelling is gericht op hun expressie in lichaamsvloeistoffen. Dit komt door de ontregeling in menselijke ziekten zoals kanker en hun mogelijke toepassing als serumbiomarkers. Echter, de analyse van miRNA expressie in serum problematisch zijn. In de meeste gevallen is de hoeveelheid serum wordt beperkt en serum bevat geringe hoeveelheden totaal RNA, waarvan slechts kleine RNAs vormen 0,4-0,5% 1. Dus de isolatie van voldoende hoeveelheden van kwaliteit RNA uit serum is een grote uitdaging voor onderzoekers vandaag. In dit technische document tonen we een werkwijze die op 400 gl menselijk serum gebruikt om voldoende RNA voor zowel DNA arrays of qPCR analyse. De eendvantages van deze werkwijze zijn de eenvoud en het vermogen om hoge kwaliteit RNA opbrengst. Het vereist geen speciale kolommen voor zuivering van kleine RNA&#39;s en maakt gebruik van algemene reagentia en hardware gevonden in gemeenschappelijke laboratoria. Onze werkwijze gebruikt een Phase Lock Gel fenol verontreiniging te elimineren terwijl tegelijkertijd de hoge kwaliteit waardoor RNA. We introduceren ook een extra stap om alle verontreinigingen verder te verwijderen tijdens de isolatie stap. Dit protocol is zeer effectief bij het isoleren van totaal RNA opbrengst tot 100 ng / ul uit serum, maar kan ook worden aangepast voor andere biologische weefsels.

Figuur 1 toont een typische UV / Vis-spectrum van RNA geïsoleerd uit serum. Uit dit profiel hebben we vastgesteld eiwit verontreinigingen bij 280 nm met fenol en organische verontreinigingen zowel 270 nm en 230 nm respectievelijk. Residu guanidinium thiocyanaat werd waargenomen bij 260 nm. Om verontreinigingen te verminderen, een aantal optimalisatie stappen gezet naar de standaard Tri-reagens RT-LS procedure.
