Extra-und intrazellulären elektrophysiologischen Ableitungen neuronaler Aktivität und synaptische Potentiale in vivo sind Stand der Technik Techniken bei der Untersuchung von ZNS neuronale Funktionen und Pathophysiologie. Spinal Integration ist entscheidend für die motorische Funktion, z. B. Bewegung des Körpers und für multimodale Sinneswahrnehmung. Präsynaptischen Hemmung ist einer kritischen Mechanismus in diesem Rechenvorgang eine angemessene Reaktionen auf Sinneseindrücke. GABAergen Synapse...

Präsynaptischen Inhibierung ist eine der stärksten inhibitorischen Mechanismen im Rückenmark. Es hemmt die exzitatorischen postsynaptischen Potentiale (EPSPs) in monosynaptisch angeregten Motoneuronen, ohne die postsynaptischen Membranpotential und die Erregbarkeit der Motoneuronen 1-3. Primär afferente Depolarisation (PAD), die durch GABA-erge axo-axonale Synapsen auf sensorische präsynaptischen Fasern induziert wird, ist der zugrunde liegende Mechanismus 4-7 (siehe auch Abbildung 1a). Diese Synapsen enthalten GABA A-und GABA-B-Rezeptoren (GABA A R-und GABA B-R). GABA A-R-Aktivität führt zu einer Erhöhung der Chlorid-Leitfähigkeit, die PAD aufgrund des lokalen Ionenverteilung hervorruft. Diese Depolarisation blockiert die Ausbreitung von Aktionspotentialen in die Axon-Terminals und reduziert ihre Stärke, die zu einer verminderten Ca 2 +-Einstrom und eine Reduktion der Transmitterfreisetzung. Aktivierung der GABA-B-Rezeptoren tut nichtt PAD beitragen, aber zu einer Verringerung der Ca 2 +-Einstrom wodurch präsynaptischen Inhibierung. Während die Aktivierung der GABA A R scheint in kurzen Zeithemmung beteiligt sind, werden GABA B-R der langfristigen Modulation 8-10 beteiligt. Neben GABA, die den größten Teil der PAD und präsynaptischen Hemmung ausmacht, könnten auch andere Sender Systeme modulieren und dazu beitragen, diesen Mechanismus 11,12. Pathologische Veränderungen in der präsynaptischen Hemmung scheint entscheidend zu sein in mehreren Krankheitszuständen wie zB periphere neuropathische Schmerzen und Entzündungen 13,14 sowie abnorme zentrale Schmerzverarbeitung 15, Verletzungen des Rückenmarks 16 und ZNS-Erkrankung mit motorischen Übererregbarkeit durch defekte GABAergen Transmission 17 vermittelt werden, 18. So Schätzung präsynaptischen Hemmung lohnt sich, experimentellen pathologischen Bedingungen auf der Ebene des Rückenmarks in vivo zu untersuchen </em&gt;. PAD entsteht Volumen durchgeführt Potentiale eine direkte Messung der präsynaptischen Inhibierung im Rückenmark. Diese Potentiale werden Hinterwurzel Potentiale (DRP) genannt und kann von Rückenmark dorsalen Wurzeln nach Stimulation von benachbarten dorsalen Wurzeln 7 gemessen werden. Erste Messungen von DRP haben bei Katzen und Frösche 19 gemeldet und wurden intensiv bei Katzen von Eccles, Schmidt und andere, in den frühen 1970er Jahren 3,4,20,21 sucht. Während in vivo-Aufnahmen von DRP bei Katzen 22 und 23 Ratten wurden weit verbreitet, Messungen in Mäusen wurden fast ausschließlich im ex vivo isolierten Rückenmark Zubereitungen 15,24 durchgeführt. Hier beschreiben wir ein Verfahren zur DRP an narkotisierten Mäusen in vivo ermöglicht eine direkte Messung der präsynaptischen Inhibierung im intakten Organismus aufzuzeichnen.

Präsynaptischen Inhibierung ist eine der stärksten inhibitorischen Mechanismen im Rückenmark. Die zugrunde liegende physiologische Mechanismus ist eine Depolarisation der primären afferenten Fasern durch GABA-erge axo-axonale Synapsen (primäre afferente Depolarisation) vermittelt. Die Stärke der primären afferenten Depolarisation durch Aufnahme der auf das Volumen durchgeführt, die Potentiale an den dorsalen Wurzel (Hinterwurzelpotentiale DRP) gemessen werden. Pathologische Veränderungen der präsynaptischen Hemmung sind in der abnormen zentralen Verarbeitung bestimmter Schmerzzuständen und in einigen Störungen des Motor Übererregbarkeit von entscheidender Bedeutung. Hier beschreiben wir ein Verfahren zum Aufzeichnen DRP in vivo in Mäusen. Die Vorbereitung des Rückenmarks Rücken Wurzeln in der narkotisierten Tier und der Aufnahmeverfahren mit Saug-Elektroden werden erläutert. Diese Methode ermöglicht die Messung GABAerge DRP und damit die Schätzung Rücken präsynaptischen Hemmung in der lebenden Maus. In Kombination mit transgenen Mausmodellen, kann DRP Aufnahme serve als ein leistungsfähiges Werkzeug, um krankheitsassoziierten Rücken Pathophysiologie zu untersuchen. In-vivo-Aufnahme hat mehrere Vorteile im Vergleich zu ex vivo isolierten Rückenmark Mittel, wie zB die Möglichkeit der gleichzeitigen Aufnahme oder Manipulation von supraspinalen Netzwerke und Induktion von DRP durch Stimulation der peripheren Nerven.

Typische DRP Spuren sind in Fig. 3 gezeigt. Die prominente Stimulationsartefakt wird üblicherweise durch eine kurze Abwärtsbiegung folgt. Danach ein langsamer, dauerhafte Auslenkung nach oben, die die DRP deutlich unterscheidbar. In einer Untergruppe von Aufnahmen sind Algan Reflexe, wie kleine Spitzen auf der Oberseite der DRP sichtbar.
