En résumé, nous avons pu démontrer que la liaison CH dans tétrahydrocarbazoles peut être facilement fonctionnalisés pour générer des produits CN-couplage dans une procédure en deux étapes. La première étape est une oxydation photocatalyzed bien connu de tétrahydrocarbazole (1) ou de ses dérivés avec de l&#39;oxygène élémentaire 17,19, ce qui donne un hydroperoxyde 2. La fonctionnalisation directe de liaisons CH est un objectif permanent, important et très en chimie organique 1. Ces transformations peuvent être très puissants afin de simplifier la synthèse en enregistrant étapes, le temps et le matériel par rapport aux méthodes classiques qui nécessitent la mise en place et le retrait de l&#39;activation ou de diriger des groupes. Par conséquent, la fonctionnalisation de liaisons CH est également attrayant pour la chimie verte 2. Dans des conditions oxydantes, deux liaisons CH ou une liaison CH et un hétéroatome-H peut être transformé en CC et des liaisons C-hétéroatome, respectivement (Figure 1) 3-9. Souvent, ces réactions de couplage oxydatif nécessitent oxydants synthétiques, des catalyseurs coûteux ou des températures élevées. Par conséquent, de nombreuses tentatives sont faites pour développer des méthodes qui utilisent des catalyseurs bon marché, les conditions bénignes et de l&#39;oxygène ou de l&#39;air comme oxydant borne 10. <img alt="Figure 1" fo:content-width = &quot;5 po&quot; src = &quot;/ files/ftp_upload/51504/51504fig1highres.jpg&quot; width = &quot;500&quot; /&gt; Figure 1. Récemment, nous avons découvert une réaction qui utilise une auto-oxydation pour former une nouvelle liaison CC de liaisons CH, sans nécessité d&#39;un catalyseur ayant une activité rédox 13,14 </sup&gt;. Remuant simplement les substrats, sous oxygène, en présence d&#39;un catalyseur acide, conduit à la formation de ces nouveaux produits. La clé de la réaction est la formation d&#39;hydroperoxydes facile intermédiaires, qui sont substitués par le second substrat 15 par catalyse acide. La réaction est toutefois limité à xanthénique et quelques composés apparentés qui sont facilement oxydés sous une atmosphère d&#39;oxygène et les produits ont des applications jusqu&#39;ici pas trouvé. Néanmoins inspiré par cette découverte, nous avons développé une méthode de couplage oxydatif lié qui utilise le principe de CH fonctionnalisation via intermédiaires peroxydes (CHIPS) pour synthétiser pharmaceutique dérivés d&#39;indole actifs 16. Indoles, en particulier une tétrahydrocarbazoles, peuvent être facilement oxydés en deux hydroperoxydes en présence d&#39;oxygène singulet 17 à 19, qui peut être généré en utilisant un sensibilisateur à la lumière visible et 20. Un hydrgroupement operoxide peut en principe servir d&#39;un groupe partant s&#39;il est activé par catalyse acide et permettre la mise en place d&#39;un nucléophile 21,22. Les hydroperoxydes sont également connus pour subir des réactions de réarrangement catalysé par un acide tel qu&#39;il est utilisé dans la synthèse industrielle de phénol à partir de cumène, le 23 procédé de Hock. Par des études d&#39;optimisation prudents, nous pourrions trouver des conditions pour favoriser la réaction de substitution souhaitée avec N-nucléophiles comme anilines 3 sur les voies de décomposition indésirables par réarrangement 16. Ici, nous décrivons cette procédure CHIPS deux étape en détail, en utilisant seulement la lumière visible, un sensibilisateur, de l&#39;oxygène et de l&#39;acide. Parmi les produits sélectionnés sont dérivés d&#39;indole 4, qui montrent une activité antivirale élevée ou inhibent le facteur de croissance endothélial vasculaire (VGF), qui peut être important pour le traitement des tumeurs de 24 à 26.

The direct functionalization of C-H bonds is an important and long standing goal in organic chemistry. Such transformations can be very powerful in order to streamline synthesis by saving steps, time and material compared to conventional methods that require the introduction and removal of activating or directing groups. Therefore, the functionalization of C-H bonds is also attractive for green chemistry. Under oxidative conditions, two C-H bonds or one C-H and one heteroatom-H bond can be transformed to C-C and C-heteroatom bonds, respectively. Often these oxidative coupling reactions require synthetic oxidants, expensive catalysts or high temperatures. Here, we describe a two-step procedure to functionalize indole derivatives, more specifically tetrahydrocarbazoles, by C-H amination using only elemental oxygen as oxidant. The reaction uses the principle of C-H functionalization via Intermediate PeroxideS (CHIPS). In the first step, a hydroperoxide is generated oxidatively using visible light, a photosensitizer and elemental oxygen. In the second step, the N-nucleophile, an aniline, is introduced by Br&#248;nsted-acid catalyzed activation of the hydroperoxide leaving group. The products of the first and second step often precipitate and can be conveniently filtered off. The synthesis of a biologically active compound is shown.
