two methods for establishing primary human endometrial stromal cells from hysterectomy specimens

Mange bestræbelser har været afsat til at etablere in vitro-cellekultur-systemer. Disse systemer er designet til at modellere et utal af in vivo processer. Cellekultursystemer opstår fra humane endometriske prøver er ingen undtagelse. Applikationer spænder fra normale cykliske fysiologiske processer til endometrie patologier såsom gynækologisk cancer, infektionssygdomme og nedsat fertilitet. Her giver vi to metoder til oprettelse primære endometrie stromaceller fra kirurgisk resektion endometrie hysterektomi prøver. Den første metode er benævnt &quot;skrabe&quot;-metoden og omfatter mekanisk skrabning ved hjælp af kirurgisk eller barberblade, mens den anden metode betegnes &quot;trypsin-metoden&quot;. Sidstnævnte metode anvender den enzymatiske aktivitet af trypsin til at fremme adskillelse af celler og primære celleudgroning. Vi illustrerer trin-for-trin metode gennem digitale billeder og mikroskopi. Vi har også provide eksempler på validering endometriale stromacellelinier via kvantitative realtids polymerasekædereaktioner (qPCR) og immunfluorescens (IF).
