Ce protocole décrit une méthode simple pour automatiquement fluorescence spécifique des tissus image embryons de poisson zèbre. Le protocole a été développé en utilisant un Zeiss Axio Observer. Z1 avec Zen logiciel Blue 2011, cependant la technique peut être adaptée en utilisant n&#39;importe quel microscope inversé avec une platine motorisée et un logiciel de contrôle de microscope qui peut effectuer carrelage pour créer des images composites. Poisson-zèbre transgénique ont été développés qui permettent la visualisation directe de l&#39;activité dans les voies, telles que les facteurs de croissance des fibroblastes 1, l&#39;acide rétinoïque et les œstrogènes 2 3, dans embryons vivants signalisation. De tels outils permettent le dépistage de substances qui perturbent les voies de signalisation (dosée comme changement de l&#39;intensité de fluorescence) ou à des composés chimiques qui modulent la signalisation d&#39;une manière spécifique d&#39;un tissu (changement de fluorescence localisation) 4. Capture d&#39;image automatisé augmente le débit d&#39;écrans chimiques considérablement 5,6. Écrans fluorescence dosage automatiquement chez le poisson zèbre en direct s&#39;appuient souvent sur des équipements coûteux, spécialisé. Soi-disant criblage à haut contenu offre l&#39;avantage de la haute résolution, l&#39;imagerie quantitative, mais au prix de l&#39;utilisation de lecteurs de plaques spécialisées équipées pour la microscopie confocale 7,8. Le but de cette méthode est d&#39;automatiquement fluorescence spécifique des tissus essai dans embryons de poissons zèbresdans une plaque à 96 puits en utilisant un microscope à épifluorescence standard. Fluorescence spécifique d&#39;un tissu peut être distingué en utilisant cette technique et peut-être une approche raisonnable pour les laboratoires qui n&#39;ont pas accès aux lecteurs de plaques spécialisées ou de l&#39;équipement de haute teneur dépistage. Dans ce protocole, nous utilisons l&#39;imagerie automatisée pour détecter récepteur des oestrogènes tissu-spécifique (ER) des agonistes chez le poisson zèbre en direct à 3 jours après la fécondation (DPF). La lignée transgénique Tg (5xERE: GFP) c262/c262 contient 5 tandem réponse aux œstrogènes séquences d&#39;ADN d&#39;éléments (ERE) en amont de la protéine fluorescente verte (GFP) 3. En l&#39;absence de ligand, RÉ sont normalement inactifs. La liaison du ligand déclenche un changement de conformation, ce qui permet de lier les récepteurs ERE ADN et régulent la transcription 9. 5xERE: poissons GFP peuvent être utilisés pour cribler des bibliothèques chimiques pour modulateurs ER et peuvent être utilisés pour cribler des échantillons d&#39;eau de l&#39;environnement pour les contaminants oestrogéniques.

Embryons de poisson zèbre constituent un outil puissant pour le dépistage à grande échelle de petites molécules. Poisson-zèbre transgénique qui exprime des protéines rapporteurs fluorescents sont fréquemment utilisés pour identifier des produits chimiques qui modulent l&#39;expression des gènes. écrans chimiques qui essai de fluorescence chez le poisson zèbre en direct s&#39;appuient souvent sur des équipements coûteux, spécialisé pour le criblage à haut contenu. Nous décrivons une procédure à l&#39;aide d&#39;un microscope à épifluorescence standard avec une platine motorisée à automatiquement l&#39;image des embryons de poisson zèbre et de détecter la fluorescence spécifique de tissu. Utilisation de poisson-zèbre transgénique pour signaler que l&#39;activité du récepteur des oestrogènes via l&#39;expression de la GFP, nous avons développé une procédure semi-automatique pour cribler des ligands du récepteur de l&#39;œstrogène qui activent le rapporteur d&#39;une manière tissu-spécifique. Dans cette vidéo, nous décrivons les procédures pour rangeant embryons de poisson zèbre à 24-48 heures après la fécondation (HPF) dans une plaque de 96 puits et ajouter de petites molécules qui se lient les récepteurs des œstrogènes. À 72-96 HPF, des images de chaque puits dela totalité de la plaque sont collectées automatiquement et manuellement pour inspecter fluorescence spécifique d&#39;un tissu. Ce protocole démontre la capacité de détecter les oestrogènes qui activent les récepteurs dans les valves cardiaques, mais pas dans le foie.

La figure 1 montre des images composites à partir de puits individuels d&#39;une plaque à 96 puits. Chaque image composite est composé de 59 images individuelles avec 5% d&#39;image de chevauchement. Notez que le poisson zèbre en direct sont orientés de façon aléatoire au sein de chaque bien, mais nous sommes en mesure de distinguer la fluorescence dans le coeur du foie.
