Au cours de la dernière décennie, les mutations pathogènes dans actine ont été découvertes dans chacun des six isoformes d&#39;actine humaines menant à une série de troubles, de myopathies de maladie coronarienne 1-7. Les processus par lesquels les mutations conduisent à l&#39;actine maladie continuent à être élucidée. aip1p dynamics are altered by the r256h mutation in actin

Des mutations dans actine provoquent une gamme de maladies humaines dues à des changements moléculaires spécifiques qui modifient souvent la fonction du cytosquelette. Dans cette étude, l&#39;imagerie de fluorescence protéines marquées par fluorescence interne totale (FRBR) microscopie est utilisée pour visualiser et quantifier les changements dans la dynamique du cytosquelette. microscopie TIRF et l&#39;utilisation de marqueurs fluorescents permet aussi de quantifier les changements dans la dynamique du cytosquelette causées par des mutations dans l&#39;actine. En utilisant cette technique, la quantification de la fonction du cytosquelette dans les cellules vivantes complète utilement dans des études in vitro de la fonction des protéines. A titre d&#39;exemple, des mutations faux-sens affectent le résidu R256 de l&#39;actine ont été identifiés dans les trois isoformes d&#39;actine humain suggère que cet acide aminé joue un rôle important dans les interactions régulatrices. Les effets de la mutation R256H actine du cytosquelette sur les mouvements ont été étudiés à l&#39;aide du modèle de la levure. La protéine, AIP1, qui est connu pour aider à cofilin dépolymérisation de l&#39;actine, étaitmarqués avec la protéine fluorescente verte (GFP) à l&#39;extrémité N-terminale et suivis in vivo en utilisant la microscopie TIRF. La vitesse de déplacement Aip1p à la fois de type sauvage et des souches mutantes a été quantifiée.
