Glycosomen zijn essentieel, dynamisch, parasiet-specifieke organellen. De processen die de biogenese, onderhoud, proliferatie en verbouwing van deze organellen regelen waarschijnlijk ook drug targets die kunnen worden benut voor therapeutische doeleinden. Ondanks het potentieel hoge overvloed van deze targets, is het gebied van glycosome biogenese achterbleef het bestuderen van gelijkaardige processen in andere organismen, voornamelijk door het ontbreken van een soepel, high-throughput systeem waarbij snelle, dynamische...

Trypanosoma brucei veroorzaakt Afrikaanse slaapziekte bij de mens en een slopende ziekte, Nagana, bij runderen. Geneesmiddelen die worden gebruikt bij de behandeling van deze ziekten zijn verouderd en uiterst giftig, vaccins beschikbaar zijn, en het potentieel voor de ontwikkeling van resistentie vereist de zoektocht naar nieuwe drug targets 1. Tijdens zijn levenscyclus, T. brucei, afwisselend een insect vector en zoogdiergastheer-; twee hosts die zeer verschillende omgevingen waarin de parasiet moet overleven presenteren. Een aantal metabolische en morfologische veranderingen optreden als de parasiet blootgesteld aan verschillende omgevingsomstandigheden. Enkele van de meest dramatische veranderingen zijn waargenomen bij enkele-membraan begrensd parasiet specifieke microlichamen, genoemd glycosomen 13. Glucosespiegels zijn relatief hoog (~ 5 mM) in het bloed en bloed parasieten (BSF) genereren ATP uitsluitend via glycolyse while mitochondriale stofwisseling wordt onderdrukt 14. In tegenstelling tot andere eukaryoten waar glycolyse optreedt in het cytoplasma, T. brucei compartmentalizes meeste glycolytische enzymen in glycosomen 14,15. De parasieten worden door de tseetseevlieg tijdens een bloedmeel genomen en ervaren een daling in glucose, dat valt te stellen niveaus binnen 15 minuten te worden ingenomen door de vlieg. Het metabolisme van insecten, procyclische vorm (PCF), parasieten is flexibeler en glucose, alsook aminozuren zoals proline, kan worden gebruikt bij de synthese van ATP 16-18. Vergelijkende proteomics studies blijkt lifecycle afhankelijke veranderingen in glycosomal en mitochondriale eiwitten met glycolytisch eiwitten toegenomen in de bloedbaan parasieten en mitochondriale eiwitten die betrokken zijn bij de TCA-cyclus en respiratoire keten 13,19. Terwijl veel studies hebben zich gericht op de verschillen tussen de BSF en PCF glycosomen, is er weinig bekend over de veranderingen in de PCF glycosomen die optreden als reactie op environmental veranderingen. In de dikke darm van de vlieg, het glucose laag is met een voorbijgaande verhoging tijdens een voeding 20. In de meeste in vitro studies, zijn PCF parasieten gekweekt in medium met glucose. Echter, recente studies hebben aangetoond dat PCF stofwisseling sterk verandert in reactie op de beschikbaarheid van 17 glucose. Aangezien glucose, proline opname en proline dehydrogenase activiteit verhogen 18. Deze verandering in het mitochondriaal metabolisme waarschijnlijk gepaard met een verandering in glycosome samenstelling en morfologie, maar dit is niet direct beoordeeld. Elektron en fluorescentie microscopie worden gemeenschappelijke technieken gebruikt om glycosome dynamiek in T. brucei 2,21-24. Deze protocollen zijn tijd en arbeidsintensief, duur en moeilijk aan te passen om real-time studies hoge doorvoer protocollen. Om deze beperking te overwinnen, een tl-organel reporter systeem used naar organellen in zoogdiercellen en gist te bestuderen is aangepast voor gebruik in T. brucei 12. Fluorescent organel reportersystemen zijn uitgebreid gebruikt in hogere eukaryoten zoals gist, planten en zoogdiercellen 25-27. In dergelijke systemen wordt een fluorescerend eiwit gefuseerd aan een aminozuursequentie die het eiwit aan specifieke organellen gericht. De afbraak en synthese van de beoogde eiwitten gemeten via fluorescentie en veranderingen in organel samenstelling worden gereflecteerd door veranderingen in cel fluorescentie. Wanneer het open leesraam van versterkte geel fluorescent eiwit (eYFP) gefuseerd aan een type II peroxisomale targeting sequentie (PTS2) 12, wordt de PTS2eYFP eiwit invoer in rijpe, import-bevoegde glycosomen en fluorescentie kan worden gevolgd via flowcytometrie. Variaties in glycosome preparaat worden gereflecteerd door veranderingen in de cellulaire fluorescentie. Dit systeem kan helpen bij resolVing de mechanismen die het milieu veroorzaakte veranderingen in glycosome samenstelling te regelen. Dit manuscript beschrijft de vorming van een glycosome reporter systeem PCF parasieten in combinatie met flowcytometrie om real-time glycosome dynamiek in levende parasieten volgen en een voorbeeld van hoe het is gebruikt om veranderingen in glycosome samenstelling volgen in reactie op verschillende omgevingen. Samengevat wordt glycosome samenstelling beïnvloed door extracellulaire glucose concentraties en passage van log-fase culturen in vers medium triggers veranderingen in glycosome samenstelling. Dit systeem kan worden aangepast om het dynamische gedrag van andere organellen in trypanosomen en andere parasieten bestuderen.

Trypanosoma brucei is een kinetoplastid parasiet die humane Afrikaanse slaapziekte (HAT), of slaapziekte, en een slopende ziekte, Nagana veroorzaakt, bij runderen 1. De parasiet afwisselend de bloedbaan van de zoogdier gastheer en de tseetseevlieg vector. De samenstelling van veel cellulaire organellen verandert in reactie op deze verschillende extracellulaire condities 2-5. Glycosomen zijn zeer gespecialiseerde peroxisomen waarin veel van de betrokken bij de glycolyse enzymen zijn gecompartimenteerd. Glycosome samenstelling verandert in een ontwikkelings-en milieuvriendelijke gereguleerde manier 4-11. Momenteel is de meest gebruikte technieken om glycosome dynamica te bestuderen zijn elektron en fluorescentiemicroscopie; technieken zijn duur, tijd en arbeidsintensief en niet gemakkelijk worden aangepast aan hoge doorvoer analyse. Om deze beperkingen, een fluorescerende-glycosome reporter systeem in te overwinnendie geel fluorescerend eiwit (eYFP) meer gefuseerd aan een peroxisomen targeting sequentie (PTS2), waarbij het ​​fusie-eiwit glycosomen 12 verbindt, is vastgesteld.
