Ursprünglich in der Mitte der 1990er Jahre entwickelt, die Laser Mikrodissektion (LCM) ermöglicht dem Benutzer, genau zu erfassen spezifische Zellen oder Zellbereiche von einem histologischen Gewebeschnitt mittels mikroskopischer Visualisierung 1, 2. Viele Studien, die molekulare Analyse von LCM gegen Kratzer Gewebe veranschaulichen den Wert der Methode 12.3. sivq-lcm protocol for the arcturusxt instrument

SIVQ-LCM ist eine neue Methode, die automatisiert und rationalisiert die traditionelle, benutzerabhängige Laser Dissektion Prozess. Es zielt darauf ab, eine fortschrittliche, schnell anpassbar Laser Dissektion Plattform-Technologie erstellen. In diesem Bericht beschreiben wir die Integration der Bildanalyse-Software ortsinvariantem Vektor Quantisierung (SIVQ) auf die ArcturusXT Instrument. Die ArcturusXT System enthält sowohl eine Infrarot-(IR) und Ultraviolett-(UV) Laser, so dass für bestimmte Zelle oder große Fläche Dissektionen. Das Hauptziel ist es, die Geschwindigkeit, Genauigkeit und Reproduzierbarkeit des Laser-Dissektion, Probendurchsatz zu erhöhen verbessern. Dieser neue Ansatz ermöglicht Mikrodissektion von tierischen und menschlichen Geweben in der Forschung und der klinischen Arbeitsabläufe.

Ein FFPE menschlichen Brustgewebeschnitt wurde für Zytokeratin AE1/AE3 mit einem Standard-IHC-Protokoll 23 immun. Nach dem Färben wurde das Gewebe auf dem Objektträger ArcturusXT Stufe angeordnet, und die SIVQ LCM-Protokoll wurde, wie oben beschrieben eingeleitet.
