De productie van bloedcellen, aangeduid hematopoiese wordt onderhouden door een kleine populatie van hematopoietische stamcellen en progenitorcellen (HSPCs). Eerder hebben we een methodologie om de 3D-architectuur van intacte BM te visualiseren met behulp van confocale fluorescentie en reflectie microscopie 3. We kenmerk uitbreiding van EGFP gemarkeerde HSPCs in de BM, maar het gebruik van slechts een enkele FP uitgesloten analyse van regeneratie op klonale niveau. Cell clusters met verschillende kleur spectra bleek aanvankelijk dicht bij het bot en vorderde gecentreerd over de tijd. Interessant, na meer dan 3 weken merg bestond macroscopische klonale clusters suggereert verspreiding van hematopoiese afgeleid van individuele HSPCs aansluitend in het merg, dan ruime verspreiding van HSPCs via de bloedbaan. Er was minder kleurendiversiteit op late tijdstippen, suggereert moeilijkheden transduceren langdurige herbevolking cellen met meerdere vectoren. Bovendien HSPCs gevormd clusters in de milt, een orgaan verantwoordelijk voor hematopoiesis in muizen. -HPSC afgeleide individuele cellen kunnen worden opgelost in de thymus, lymfeklieren, milt, lever, long, hart, huid, spier, vetweefsel en nieren van. De 3D-beelden kan kwalitatief en kwantitatief worden beoordeeld om de verdeling van cellen met minimale verstoringen van de weefsels waarderen. Tot slot, we illustrerend de haalbaarheid van live-dynamische studies in 4D door het combineren van resonante scannen multifoton en confocale time-lapse imaging. We ontwikkeld en gevalideerd een fluorescerende markering methodologie voor klonale volgen van hematopoietische stamcellen en voorlopercellen (HSPCs) met een hoge ruimtelijke en temporele resolutie te bestuderen in vivo hematopoiesis behulp van de muizen beenmergtransplantatie experimenteel model. Genetische combinatorische markering met behulp van lentivirale vectoren die coderen voor fluorescerende eiwitten (KP&#39;s) geactiveerd lot van de cel in kaart brengen door middel van geavanceerde microscopie beeldvorming. Vectoren die coderen voor vijf verschillende KP&#39;s: Cerulean, EGFP, Venus, tdTomato en mCherry werden gebruikt om gelijktijdig transduce HSPCs, het creëren van een divers palet van kleur gemarkeerd cellen. Beeldvorming met behulp van confocale / twee-foton hybride microscopie maakt gelijktijdig hoge resolutie beoordeling van unieke gemarkeerde cellen en hun nakomelingen in combinatie met structurele componenten van de weefsels. Volumetrische analyse over grote gebieden bekend dat spectraal gecodeerde HSPC-afgeleide cellen niet-invasieve wijze verschillende intacte weefsels, waaronder het beenmerg kan worden gedetecteerd (BM), Voor langere perioden na de transplantatie. Levende onderzoeken gecombineerd video-rate multiphoton enconfocale time-lapse imaging in 4D tonen de mogelijkheid van dynamische cellulaire en klonale volgen op een kwantitatieve wijze.

Whole-mount 3D confocale / 2-foton microscopie reconstructies van het borstbeen beenmerg tijdsverloop bleek de innesteling en uitbreiding van getransplanteerde co-opnamen van 5 bps in een patroon met opvallende kenmerken: klonen bleek duidelijk afgebakend, homogeen gemarkeerd met breed palet aan kleuren in eerste instantie en vooruitgang tijd preferentieel bevatten cellen van voornamelijk een kleur.
