Die Produktion von Blutzellen, genannt Hämatopoese durch eine kleine Population von hämatopoetischen Stamm-und Vorläuferzellen (HSPCs) gehalten. Bisher haben wir eine Methodik, um die 3D-Architektur von intakten BM mit Hilfe der konfokalen Fluoreszenzmikroskopie und Reflexion 3 zu visualisieren. Wir gekennzeichnet Ausbau der EGFP-markierte HSPCs in der BM, aber die Verwendung von nur einer einzigen FP ausgeschlossen Analyse der Regeneration bei einer klonalen Ebene. Erst vor kurzem haben wir den Vorteil des lentiviralen Gene Ontology (Lego)-Vektoren, die konstitutiv fluorescent Proteine ​​(RP), um effizient zu markieren Zellen 4,5. Es war weniger Farbvielfalt am späten Zeitpunkten, was auf Schwierigkeiten bei der Wandlerlangfristige Bestandsauffüllung Zellen mit mehreren Vektoren. Zusätzlich gebildet HSPCs Cluster in der Milz, ein Organ auch die Hämatopoese bei Mäusen verantwortlich ist. HPSC abgeleiteten einzelnen Zellen konnten in Thymus als auch gelöst werden, Lymphknoten, Milz, Leber, Lunge, Herz, Haut, Skelettmuskel, Fettgewebe und Niere. Die 3D-Bilder können qualitativ und quantitativ bewertet werden, um die Verteilung der Zellen mit minimalen Störungen der Gewebe zu schätzen. Schließlich zeigen wird die Machbarkeit der Live-dynamische Untersuchungen in 4D durch die Kombination von Resonanz Scannen Multi und konfokaler Zeitraffer-Bildgebung. in vivo clonal tracking of hematopoietic stem and progenitor cells marked by five fluorescent proteins using confocal and multiphoton microscopy
