A produção de células sanguíneas, denominado hematopoiese, é mantida por uma pequena população de células estaminais e progenitoras hematopoéticas (hspCs). Estas células residir dentro da medula óssea (BM) em um nicho microambiental complexo que consiste em osteoblastos, células do estroma, tecido adiposo e estruturas vasculares, todos implicados no controlo da auto-renovação e diferenciação 1,2. Como intacta BM tem sido tradicionalmente inacessível para observações diretas, as interações entre HSPC e seu microambiente permanece em grande parte não caracterizada in vivo. Anteriormente, nós estabelecemos uma metodologia para visualizar a arquitetura 3D da BM intacto usando microscopia de fluorescência confocal e reflexão 3. Foram caracterizados expansão de hspCs EGFP-marcados na MC, mas o uso de apenas um único FP impedida análise de regeneração a um nível clonal. Muito recentemente, aproveitou a Lentivirus Gene Ontology (Lego) vetores expressando constitutivamente flproteínas uorescent (FPS) para marcar de forma eficiente as células 4,5. A co-transdução de HSPC com três ou cinco vectores gera uma paleta de cores diversas combinatórias, permitindo a localização de vários clones de HSPC individuais. Marcado HSPC combinado com nova tecnologia de imagem permitido traçar homing indivíduo HSPC e enxerto na BM de ratos irradiados. Vetores Lego foram usadas para marcar HSPC com 3 ou 5 fps diferentes (de Cerulean, eGFP, Vênus, tdTomato e mCherry) e seguir o seu enxerto ao longo do tempo na BM por microscopia confocal e dois fótons, que permite a visualização nítida de ossos e outros estruturas de matriz, e a relação de clones HSPC de componentes do seu microambiente. HSPC foram purificados como a linhagem de células negativas de camundongos C57BL / 6 BM, transduzidas com vetores de Lego, e infundidas por injeção na veia da cauda em ratos congênitas ablated-mielo. Através do monitoramento de grandes volumes de tecido esterno BM visualizamos o enxerto intra-ósseo que ocorre no inícioBM regeneração. Grupos de células com espectros cor diversificada, parece, inicialmente, em estreita proximidade com o osso e progrediu centralmente ao longo do tempo. Curiosamente, após mais de 3 semanas, a medula consistiu de aglomerados clonais macroscópicas, sugerindo propagação da hematopoiese derivado de hspCs individuais contiguamente na medula, em vez de difusão generalizada de hspCs através da corrente sanguínea. Havia menos diversidade de cores em pontos de tempo de atraso, sugerindo dificuldade na transdução de longo prazo repovoar células com múltiplos vetores. Além disso, hspCs aglomerados formados no baço, um órgão também responsável para a hematopoiese em ratinhos. Células individuais derivadas de HPSC poderia ser resolvido no timo, nódulos linfáticos, baço, fígado, pulmão, coração, pele, músculo esquelético, tecido adiposo, e renal bem. As imagens em 3D pode ser avaliada qualitativamente e quantitativamente a apreciar a distribuição de células com perturbações mínimas dos tecidos. Por fim, ilustramosd a viabilidade de estudos dinâmicos ao vivo em 4D, combinando multiphoton digitalização ressonante e confocal de imagem time-lapse. Esta metodologia permite que não-invasivo de alta resolução, multidimensional célula-destino traçado do espectro marcados populações de células em sua arquitetura intacta 3D, fornecendo uma ferramenta poderosa no estudo da regeneração de tecidos e patologia.

Nós desenvolvida e validada uma metodologia de marcação para rastreamento clonal de células tronco e progenitoras hematopoéticas (hspCs) com alta resolução espacial e temporal para estudar in vivo hematopoiese usando o transplante de medula óssea de camundongos modelo experimental fluorescente. Marcação usando lentivirais que codificam proteínas fluorescentes (FPS) combinatória genética permitem o mapeamento do destino da célula através de imagens de microscopia avançada. Vetores que codificam cinco PQ diferentes: Cerulean, EGFP, Vênus, tdTomato e mCherry foram utilizados para a transdução simultaneamente hspCs, criando uma paleta variada de cores de células marcadas. Imagem por microscopia confocal híbrido / two-photon permite a avaliação alta resolução simultânea de células marcadas com exclusividade e seus descendentes em conjunto com os componentes estruturais dos tecidos. Análise volumétrica ao longo de grandes áreas revelam que as células derivadas de HSPC espectralmente codificados podem ser detectados de forma não invasiva, em vários tecidos intactos, incluindo a medula óssea (BM), Por extensos períodos de tempo a seguir ao transplante. Estudos ao vivo combinando multiphoton taxa de vídeo e confocal de imagem time-lapse em 4D demonstrar a possibilidade de rastreamento de celular e dinâmica clonal de uma forma quantitativa.
