Ikke desto mindre mærkbare artsspecifikke forskelle blandt remodeling komplekser, der findes, som følge af gevinst eller tab af artsspecifikke subunits, gevinst eller tab af artsspecifikke domæner af konserverede underenheder, og sekvensvariabilitet inden konserverede domæner af bevarede underenheder. Sådanne forskelle kan i princippet være drevet af behovet for højere eukaryote celler til at tilpasse sig nye molekylære og cellulære miljøer. Således at forstå, hvordan underenheder af højere eukaryote remodeling komplekser bidrager til nukleosom remodeling processen er værdifuld, fordi det ikke kun kaster lys over de grundlæggende mekanismer i ATP-afhængig kromatin remodellering proces, men også kan give værdifuld indsigt i de mekanismer, som kromatin struktur og genekspression i highendes eukaryoter reguleres. Hidtil har der kun været begrænset strukturelle og funktionelle studier af multi-subunit pattedyr kromatin remodeling komplekser, dels på grund af de vanskeligheder med at opnå biokemisk definerede kromatin remodeling komplekser og subcomplexes. Vi har delvist omgået disse vanskeligheder med de procedurer, der er beskrevet nedenfor, hvor immunaffinitetsoprensning anvendes til at fremstille intakte INO80 eller INO80 subcomplexes fra humane celler stabilt udtrykker N-terminalt FLAG epitopmærket vildtype eller mutant versioner af Ino80 5-7 (figur 1) . For at opnå intakte INO80 komplekser fra humane celler, er Flp-medieret rekombination anvendes til at generere transgene HEK293 cellelinjer, der stabilt udtrykker FLAG epitop-tagget cDNA&#39;er, der koder underenheder af INO80 kompleks 8-10. Da overekspression af INO80 underenheder kan være noget giftigt, er det nødvendigt at isolere og opretholde klonale cellelinjer under selektiv cold at sikre en stabil transgenekspression i løbet af de mange passager, der er nødvendige til ekspansion af store cellekulturer. For at opnå mindre INO80 subcomplexes, der kun indeholder en delmængde af underenheder, har vi med succes anvendt to fremgangsmåder (figur 2A, B). I den første, vi genererer HEK293 Flp-In-cellelinier stabilt udtrykker mutant versioner af Ino80 der mangler domæner, der kræves for samspil med specifikke underenheder 5. Alternativt er siRNA-medieret knockdown anvendes til at udtømme den ønskede subunit fra celler, der udtrykker en passende FLAG-mærkede INO80 underenhed (upublicerede data). generation and purification of human ino80 chromatin remodeling complexes and subcomplexes

INO80 kromatin remodeling komplekser regulerer nukleosom dynamik og DNA tilgængelighed ved at katalysere ATP-afhængig nukleosom remodeling.
