Transplantation av fasta organ såsom hud, hjärta och njurar är nu ett standardförfarande i medicinsk praxis i hela världen 1. Framgångsrikt transplanterade organ kan avvisas av aktivering av mottagarens immunsystem, som erkänner de stora histokompatibilitetsantigener av donatorn. Därför transplanterade patienter behöver behandling med immunosuppressiva läkemedel 2. Allogen hud transplantation på möss etablerades av Medawar och kollegor i 1955 och var till hjälp för att identifiera de riktade molekylerna senare beskrivs som MHC (MHC) klass I och II. Sedan dess har hudtransplantation modellen kontinuerligt modifierats och anpassats för att studera betydelsen av T-cell-undergrupper och relevansen av kemiska och biologiska intervention i trycka avstötning av transplantat 2-4. Hud från örat och bålen är svårare att förbereda och är mer mottagliga för hypoxi och nekros än tail-skin 5; docktransplantationsförfarandet är likartad. Den adoptiv överföring av Vα2Vβ8-TcR-specifika T-celler har använts för att etablera en kasse modell i immundefekta C57BL / 6 RAG2 - / - möss som transplanterats med IA bm12 hud. Genetiska skillnader mellan givare och mottagare påverkar resultatet av transplantatacceptans och avvisande. Det finns olika typer av transplantationer: auto är transplantationer från den mottagande individen själv; syngrafts och allografter är transplantationer från genetiskt identiska och genetiskt obesläktade individer respektive. Godkännande av olika transplantationer allogen organ har visats genom kemisk och biologisk ingripande i patienter och musmodeller 11,3,4. I en grundläggande tillvägagångssätt, anti-CD3-antikropp-behandlade C57BL / 6 möss uppvisade förlängd överlevnad av IA bm12 tail-skin (opublicerade data). Utarmning av CD4 + och CD8 + T-celler före transplantation i mottagarländerna möss resulterade i godkännande av MHC klass I och II inkompatibla transplantat (rev. i 12). Intressant, avstötning av hudtransplantation beror på närvaron av CD4 + </sup&gt; T-celler (rev. i 12). I denna modell kan rikta specifika interaktioner mellan olika immunceller genom att blockera samstimulerande molekyler med antikroppar eller undertryckande med regulatoriska T-celler inducerar tolerans (opublicerade data). Faktum är att blockera både CD40 och CD28 ledde till långvarig hud allograft tolerans 13,14. Svans hudtransplantation är lätt att utföra och lätt att övervaka jämfört med transplantation av andra organ. Dessutom svans-hud transplantationer är lätt att förbereda och är mindre känsliga för ischemi än andra hud vävnader. I motsats till injicerade bedövningsmedel, användning av narkosgasen (isofluoran) vid transplantation förkortar både förfarandet och mottagare återhämtningstid. Curling av svansen-hud transplantation, vilket kan leda till ofullständig sårläkning och transplantatavstötning, förhindras genom tillämpning av vävnadslim. Dessutom IA bm12 tail-hudtransplantation modell aktiverar CD4 uteslutande <sup&gt; + T-celler i både immunkompetenta och immundefekta möss (med samma genetiska bakgrund) att underlätta tolkningen av resultaten. Detta protokoll beskriver en pålitlig, reproducerbar och lätt övervakas musmodell som möjliggör kemisk och biologisk ingripande. Modellen är avsedd för undersökning av avstötning och toleransinduktion av svans-hudtransplantat.

The study of T cell responses and their consequences during allo-antigen recognition requires a model that enables one to distinguish between donor and host T cells, to easily monitor the graft, and to adapt the system in order to answer different immunological questions. Medawar and colleagues established allogeneic tail-skin transplantation in mice in 1955. Since then, the skin transplantation model has been continuously modified and adapted to answer specific questions. The use of tail-skin renders this model easy to score for graft rejection, requires neither extensive preparation nor deep anesthesia, is applicable to animals of all genetic background, discourages ischemic necrosis, and permits chemical and biological intervention. 
In general, both CD4+ and CD8+ allogeneic T cells are responsible for the rejection of allografts since they recognize mismatched major histocompatibility antigens from different mouse strains. Several models have been described for activating allogeneic T cells in skin-transplanted mice. The identification of major histocompatibility complex (MHC) class I and II molecules in different mouse strains including C57BL/6 mice was an important step toward understanding and studying T cell-mediated alloresponses. In the tail-skin transplantation model described here, a three-point mutation (I-Abm12) in the antigen-presenting groove of the MHC-class II (I-Ab) molecule is sufficient to induce strong allogeneic CD4+ T cell activation in C57BL/6 mice. Skin grafts from I-Abm12 mice on C57BL/6 mice are rejected within 12-15 days, while syngeneic grafts are accepted for up to 100 days. The absence of T cells (CD3-/- and Rag2-/- mice) allows skin graft acceptance up to 100 days, which can be overcome by transferring 2 x 104 wild type or transgenic T cells. Adoptively transferred T cells proliferate and produce IFN-&#947; in I-Abm12-transplanted Rag2-/- mice.
