fluorescence imaging with one-nanometer accuracy (fiona)

Fluoreszenz-Imaging mit ein-Nanometer-Genauigkeit (FIONA) ist eine einfache, aber nützliche Technik zur Lokalisierung von einzelnen Fluorophore mit Nanometer-Präzision in der xy-Ebene. Hier eine Zusammenfassung der FIONA Technik wird berichtet, und Beispiele der Forschung, die durchgeführt wurde unter Verwendung Fiona kurz beschrieben. Erstens, wie man für FIONA Experimente, dh ein Totalreflexions-Fluoreszenzmikroskopie (TIRFM), mit Details über die Ausrichtung der Optik setzen die erforderliche Ausrüstung, beschrieben. Dann, wie man auf die Lokalisierung immobilisiert Cy3-DNA einzelner Moleküle mit geeigneten Protokollen, gefolgt von der Nutzung von FIONA um die 36 nm Schrittweite von einer einzigen abgeschnitten Myosin Va Motor mit einem Quantenpunkt markiert messen Durchführung einer einfachen FIONA Experiment veranschaulicht. Schließlich jüngsten Bemühungen um die Anwendung FIONA zu dicken Proben erstrecken wird berichtet. Es wird gezeigt, dass mit einem Wasserimmersionsobjektiv und Quantenpunkte getränkt tief in Sol-Gele und Kaninchenauge Hornhäute (&gt;200 um) Lokalisierungsgenauigkeit von 2-3 nm erreicht werden.

Ein typisches Ziel-Typ TIRFM Aufbau ist in Abbildung 3 dargestellt. Zuerst wurde auf der Oberfläche immobilisierten Cy3-DNA Probe abgebildet. Ein typisches Bild ist in 4a gezeigt. Das Bild wurde mit 0,5 Sekunden Belichtungszeit aufgenommen, mit EM Verstärkung = 50 und CCD-Empfindlichkeit = 12.13 für die Kamera.
