Vår kunnskap om celle ultrastructure kommer hovedsakelig fra elektronmikroskopi, som kan løse detaljer i størrelsesorden noen få nanometer en. Plantevev er av spesiell interesse på grunn av tilstedeværelsen av karbonisert mellomrom og vannfylte vakuoler som hindrer isfritt frysing av vann. I tillegg er prosessen med kjemisk fiksering særlig lenge i planter på grunn av celleveggene som hindrer inntrengning av kjemikaliene til dyp i vev. Plantemateriale er derfor particsig utfordrende, men denne protokollen er pålitelig og produserer prøver av høyeste kvalitet.

Resultatene som presenteres nedenfor er innhentet ved hjelp av en Wohlwend Compact 02 for HPF (figur 1A). En stor fordel med dette instrumentet er den enkle bruken av prøven bærere og dets eiere. Ved bruk av andre instrumenter, anbefaler McDonald at to brukere skal utføre prøveopparbeidelse og HPF, en forbereder prøvene mens den andre gjør frysing og overføring til FS cryovials ni. Tandem High-pressure Freezing and Quick Freeze Substitution of Plant Tissues for Transmission Electron Microscopy

Obtaining high-quality transmission electron microscopy images is challenging, especially in the case of plant cells, which have abundant large water-filled vacuoles and aerated spaces. Tandem high-pressure freezing and quick freeze substitution greatly reduce preparation time of plant samples for TEM while producing samples with excellent ultrastructural preservation.
