Behovet av att genomföra globala analyser av proteinbiokemi och bindande aktivitet in vivo har resulterat i utvecklingen av nya metoder för profilering protein-proteininteraktioner (protonpumpshämmare) och posttranslationella modifieringar av hela proteom 1,3-8. Protein mikroarrayer tillverkas som funktionellt protein mikroarrayer använder fullängds funktionella proteiner 4-6,8,9 eller analytiska protein mikroarrayer innehållande antikroppar 10,11. De är konstruerade för att innehålla en hög densitet av proteiner anordnade på objektglas med en mängd yta kemier för att underlätta en mängd olika experimentella villkor som krävs för att genomföra omfattande biokemiska analyser 12. Syftet med detta protokoll är att visa lämplig användning av funktionellt protein microarrays för att detektera protein-proteininteraktioner. Denna applikation gör det möjligt för hög genomströmning parallell biokemisk analys av proteinbindningsaktiviteter med en högrenat analyt (protein) av intresse. En C-terminalt (karboxiterminala) taggade V5-fusionsbetesprotein av intresse framställs från en hög-kopia plasmid i en jäststam optimerad för proteinrening. C-terminala märkning säkerställer att fullängdsproteinet har översatts. Det protein som används i denna studie är Tda1-V5-fusionsproteinkinas, som renas med användning av nickel affinitetsharts via en His6X tagg. Den Tda1-V5-fusions byggt renas genom seriell eluering med användning av en imidazol-gradient för att eluera den högst anrikade fraktionen för användning i analysen.
