Hier presenteren we een uniek systeem om chronologisch te analyseren muis foetale lever terminal erytropoëse. Door de toepassing van verschillende kweekomstandigheden, we met succes ontleed terminal erytropoëse in de vroege en late stadia. Dit is bijzonder belangrijk om de mechanismen van genen met meerdere functies bepalen. Bijvoorbeeld, Rac GTPases spelen een belangrijke rol in verschillende stadia van terminal erythropoiese. Erytropoëse is het proces van differentiatie van multipotente hematopoëtische stamcellen erytrocyten rijpen. Deze stapsgewijze werkwijze omvat de vorming van erytroïde burst vormende eenheden (BFU-E), de snel delende erytroïde kolonievormende eenheden (CFU-E) en morfologisch herkenbare erytroblasten 1,2. Terminal erythropoiesis van CFU-E stamcellen betreft sequentiële erytropoëtine-afhankelijke en onafhankelijke fasen 2,3. In het vroege stadium van terminal erytropoëse, Erytropoëtine (Epo) bindt aan zijn receptor op het celoppervlak en induceert een reeks stroomafwaartse signalering pathways die cel apoptose te voorkomen en snelle celdelingen en genexpressie 1,4. In de late fase van de terminal erytropoëse, erytroblasten ondergaan terminal celcyclus exit, chromatine en nucleus condensatie, en extrusie van de sterk gecondenseerde kernen 5. Ons begrip van de terminalerytropoëse is sterk verbeterd in de afgelopen decennia, dat is grotendeels te danken aan het succesvolle gebruik van verschillende in vitro en in vivo muismodellen 6-9. Van deze modellen in vitro kweek van muizen foetale lever erythroblasten biedt vele voordelen, waaronder het gemak van cel zuivering, snelle proliferatie en differentiatie, en een dichter imiteren menselijke erytropoëse 10,11. In dit systeem kunnen grote aantallen erythroïde progenitorcellen uit muizen foetale levers gemakkelijk worden geïsoleerd door het enkele stap zuivering van TER119 (marker voor het rijpe erytroïde cellen) negatief erytroblasten. Tijdens de tweedaagse kweek van de erytroblasten, kan de differentiatie van deze cellen worden door een flowcytometrische analyse op basis van oppervlakte-expressie van de transferrine receptor (CD71) en TER119 antigen 12. Bovendien kan enucleatie van terminaal differentiatie erytroblasten worden gedetecteerd door een DNA maker (Hoechst 33342) 13. Ook het gezuiverde voorlopercellen kunnen genetisch gemodificeerd door exogene expressie van cDNA&#39;s of kleine hairpin RNAs (shRNAs) voor de genen van belang, waarvan de mechanistische studies van de functies van genexpressie op erytropoëse 11,13,14 vergemakkelijkt. Anderzijds kan de snelle celgroei tempo een tweesnijdend zwaard zijn, aangezien het moeilijk is om genfuncties karakteriseren in verschillende stadia van terminal erythropoiese. In de meeste gevallen is het moeilijk te bepalen of een specifieke genfuncties in het vroege stadium van terminal erytropoëse sinds de tijd dat de cDNA&#39;s of shRNAs tot expressie, de cellen reeds vroeg stadium gepasseerd. Om dit probleem op te lossen, ontwikkelden we een uniek systeem voor de vroege en late stadia van de terminal erytropoëse ontleden. Bij de vroege fase van terminal erytropoëse genetisch gemodificeerde TER119 negatieve erythroblasten werden gekweekt in Epo-medium maar bevat stamcel factor (SCF), IL-6 en FLT3ligand hun progenitor status te behouden en laat de getransduceerde cDNAs of shRNA expressie 13. De cellen werden veranderd naar Epo bevattende medium na 12 uur tot celproliferatie en differentiatie induceren. Op deze manier, wanneer de cellen beginnen te differentiëren, de getransduceerde cDNA&#39;s of shRNAs reeds expressie. Voor het late stadium van terminal erytropoëse, TER119 negatieve erythroblasten werden in Epo bevattend medium onmiddellijk na transductie. Daarom kan men analyseren de functies van de genen van belang in het late stadium van terminal erythropoiese. Samengevat zou een brede toepassing van deze regeling te ontleden genfuncties in verschillende stadia van terminal erythropoiese.

Erytropoëse omvat een dynamisch proces dat begint met erytroïde burst vormende eenheden (BFU-E), gevolgd door snel delende erytroïde kolonievormende eenheden (CFU-E). Na CFU-Es, cellen zijn morfologisch herkenbare en algemeen genoemd terminal erytroblasten. Een van de uitdagingen voor de studie van terminal erytropoëse is het ontbreken van experimentele benaderingen genfuncties ontleden chronologisch wijze. In dit protocol beschrijven we een unieke strategie genfuncties bepalen de vroege en late stadia van terminal erythropoiese. In dit systeem, muis foetale lever TER119 (volwassen erytroïde-cel marker) negatief erytroblasten werden gezuiverd en getransduceerd met exogene expressie van cDNA&#39;s of kleine hairpin RNAs (shRNAs) voor de genen van belang. De cellen werden vervolgens gekweekt in medium dat dan erythropoietine (Epo) hun voorlopercellen stadium behouden gedurende 12 uur terwijl het exogene cDNA&#39;s of shRNAs groeifactorente drukken. De cellen werden veranderd naar Epo medium na 12 uur tot celdifferentiatie en proliferatie induceren terwijl het exogene genetische materiaal al werden uitgedrukt. Dit protocol maakt analyse van genfuncties in het beginstadium van terminal erythropoiese. Te laat stadium terminal erytropoëse te bestuderen, werden de cellen onmiddellijk gekweekt in Epo medium na transductie. Zo werden de cellen reeds gedifferentieerd dat late stadium van terminal erytropoëse wanneer de getransduceerde genetisch materiaal werden uitgedrukt. Wij raden een algemene toepassing van deze strategie die zou helpen gedetailleerde gen functies in de verschillende stadia van de terminal erytropoëse begrijpen.

Figuur 1 schetst de experimentele strategieën. Het protocol bestaat uit twee onafhankelijke voorwaarden voor targeting naar de functies van de signaalmoleculen in de vroege en late stadia van terminal erythropoiese. TER119 negatieve foetale lever erythroblasten werden gezuiverd uit E13.5 muis foetus.
