Historisch,verschillende experimentele methoden zijn gebruikt, waaronder injectie van antigen voorafgaand aan de injectie van kleurstof, injectie van kleurstof vóór injectie van antigeen en gelijktijdige injectie van kleurstof en antigeen. Intraveneuze toediening kleurstof als middel voor het meten van allergische reacties is een veelzijdig assay als het kan worden gebruikt voor het meten van actieve, passieve en reverse passieve reacties 5,9. Tal van kleurstoffen zijn gebruikt om allergische reacties, waaronder trypaanblauw, pontamine Sky Blue, Evans Blue, Geigy Blue 536 en India Ink 5,6,9 beoordelen. Een oplossing van 0,5% Evans Blue momenteel standaard kleurstof voor het meten van allergische reacties in de huid. De anafylactische reactie op de uitdaging is van voorbijgaande aard; maximale intensiteit wordt binnen 10 - 15 minuten kleurstof injectie en geen reactie is zichtbaar als kleurstof wordt toegediend meer dan 30 minuten na provocatie, ongeacht de gebruikte diersoorten 9. Kwantificering van kleurstof extravasatie was oorspronkelijkly verkregen door het meten wheal formaat zoals aangegeven door de blauwe kleurstof 7-9. Bovendien kan tellingen van degranulated mestcellen worden gekwantificeerd door excisie huidweefsel van de plaats van de reactie en kleuring met toluïdineblauw 7. Allergische reacties het gevolg zijn van de activatie van mestcellen en basofielen en de daaropvolgende afgifte van vasoactieve en pro-inflammatoire mediatoren. Blootstelling aan een allergeen in een gesensibiliseerd individu kan leiden tot klinische symptomen die variëren van lichte roodheid tot levensbedreigende anafylaxie. In het laboratorium zijn verschillende diermodellen ontwikkeld om de mechanismen die allergische reacties begrijpen. Hierin beschrijven we een gedetailleerde methode voor het meten van veranderingen in vasculaire permeabiliteit gelokaliseerde allergische reacties te kwantificeren. De plaatselijke anafylaxie test werd voor het eerst gemeld in de jaren 1920, en is een aangepaste versie van de techniek gepubliceerd door Kojima et al. in 2007 1. Bij deze assay muizen gesensibiliseerd met OVA uitgedaagd in de linker oor met voertuig en in het rechter oor met OVA. Dit wordt gevolgd door een intraveneuze injectie van Evans Blue kleurstof. Tien minuten na het injecteren van Evans Blue, wordt het dier gedood en de kleurstof die in extravasatiede oren wordt een nacht geëxtraheerd in formamide. De absorptie van de geëxtraheerde kleurstof wordt vervolgens gekwantificeerd met een spectrofotometer. Dieren die de test met succes hebben ondergaan zal de huid en ogen die blauw lijken te hebben. PBS gesensibiliseerde dieren mag niet reageren op PBS of OVA uitdaging, dus beide oren moet wit (Figuur 1A) blijven.
