La generación de cardiomiocitos de hPSCs, incluyendo células madre y hiPSC, puede funcionar como un modelo in vitro de los procesos de desarrollo cardíacas humanas muy tempranas, proporcionando información sobre las etapas de otra manera no accesibles para los estudios mecanicistas. Este sistema modelo ofrece oportunidades únicas para estudiar los mecanismos moleculares que controlan el compromiso de linaje cardiaco y la especificación del destino celular. En los últimos años, la capacidad de generar eficazmente células cardiomiogénico de hPSCs ha mejorado en gran medida 1-15. La elección de los marcadores, y sus correspondientes anticuerpos, que se utiliza para evaluar la pureza de in vitro cardiomyogenesis varía entre los informes. TNNT2 se ha considerado un indicador de las células comprometidas con el destino cardiomiogénico y se utiliza habitualmente para evaluar la eficacia de los protocolos de diferenciación cardíacas. Sin embargo, TNNT2 también se expresa en el músculo esquelético durante el pollo y rata desarrollo temprano 16,17 y está presente en el músculo liso humano 18 </sup&gt;. Por lo tanto, TNNT2 no es necesariamente un marcador específico de cardiomyogenesis humana in vitro. MLC2v y MLC2a se utilizan habitualmente como marcadores indirectos de subtipos ventriculares y auriculares, respectivamente. Sin embargo, los retos con confiar en MLC2v y MLC2a para determinar el subtipo de cardiomiocitos en el contexto de la diferenciación in vitro surgen del hecho de que estos productos génicos no se puede restringir a una cámara específica largo del desarrollo cardíaco, del tubo de corazón a través de adulto. En el roedor bucle corazón, MLC2a ARNm es predominante en la / zona de tracto de entrada auricular y MLC2v ARNm es predominante en las regiones del tracto ventricular / flujo de salida. En el corazón de bucle, co-expresión de MLC2a y MLC2v ARNm se observó en el tracto de entrada, canal atrioventricular y el tracto de salida 19,20. A los 3 días después del nacimiento, MLC2v ARNm está restringida al ventrículo y por 10 días después del nacimiento, MLC2a se limita a las aurículas en el corazón de rata neonatal 19. Por lo tanto, la interpretaciónde los datos relativos a la eficiencia y la identidad de subtipo cardiomyogenesis no sólo deben considerar la presencia y cantidad de los niveles de marcadores de referencia, pero deben tener en cuenta la etapa de desarrollo (s) a la que los puntos de tiempo de la diferenciación que se analizan corresponden. Esto es especialmente importante teniendo en cuenta que la etapa de maduración de las células cardiomiogénico generados por diferenciación in vitro de hPSCs se asemeja más estrechamente a los del desarrollo fetal / embrionario 21-25. Por lo tanto, basándose en la expresión espacial de un marcador en el corazón postnatal puede no ser apropiado para la evaluación de células derivadas de HPSC, al menos en algunos casos. En un esfuerzo por facilitar el desarrollo de criterios más específicos para la definición de la identidad de los cardiomiocitos in vitro, TNNI3 se considera que es un marcador útil para evaluar cardiomyogenesis in vitro, ya que se limita al músculo cardiaco a lo largo de la embriogénesis en el pollo y pez cebra 15,20y está ausente en el músculo esquelético fetal humano 26. Mientras TNNI1 está presente en el corazón fetal humano, TNNI3 es la única isoforma TNNI presente en el corazón adulto normal 27,28. En cuanto a la identidad subtipo de cardiomiocitos, Irx4 29-31 es un marcador informativo de las células con un destino ventricular. En el nivel de proteína, Irx4 ha demostrado recientemente ser restringido al ventrículo del corazón del tubo lineal a través de etapas neonatales en el ratón 32. En consecuencia, se describen los protocolos de tinción optimizados para el análisis de TNNI3 y Irx4 por citometría de flujo. Para nuestro conocimiento, esta es la primera descripción de un método para la tinción basada en anticuerpos eficiente y análisis de los niveles Irx4 en cardiomiocitos humanos por citometría de flujo.
