Met de hier beschreven werkwijzen kunnen hippocampale organotypische slice cultures worden gebruikt als een veelzijdige tool kunststof veranderingen in hersenweefsel bestuderen na beschadiging. Terwijl organotypische slice cultures in het verleden gebruikt voor het bestuderen van neuronale reacties na ischemie 6, 7 de nieuwe aspect van de gelijktijdige onderzoek van vasculaire veranderingen verbetert aanzienlijk de mogelijke toepassingen van deze methode. De inductie van OGD of hypoxie kan met vrij eenvoudige...

Het centrale zenuwstelsel is bijzonder gevoelig voor verlies of vermindering van zuurstof en glucose levering door de vasculatuur. Zelfs een vrij korte onderbreking van de bloedtoevoer naar de hersenen kan een permanent verlies van functie van het betreffende hersengebieden die tot de typische beroerte syndromen veroorzaken. Naast de neuronaal verlies in de primaire getroffen gebieden, is er meestal extra vertraagde neuronale verlies door secundaire schade. Helaas heden geen neuroprotectieve behandeling voor de vermindering van secundaire neuronale dood beschikbaar 1. Onderzoeksinspanningen voor het bestuderen van de mechanismen van secundaire schade afhankelijk van het gebruik van dierlijke modellen van cerebrale ischemie, zoals midden cerebrale slagader occlusie en diverse trombotische occlusie technieken (voor een recent overzicht zie 2). Parallel, ook als gevolg van beperkingen en ethische bezwaren met gebruik van diermodellen, organotypische slice cultuur van verschillende CNS weefsels gebruikt dat de stu toestaandy van neuronale reacties op verschillende soorten verwondingen 3-5. Voor het bestuderen van de neuronale reactie onder omstandigheden die ischemische hersenschade na te bootsen, is het modelsysteem zuurstof glucosedeprivatie (OGD) ontwikkeld. In dit model worden slice cultures tijdelijk blootgesteld aan een medium dat glucose mist en is geëquilibreerd met stikstofgas in afwezigheid van zuurstof. Met een dergelijke behandeling is het mogelijk om neuronaal letsel en schade die vrij vergelijkbaar met die waargenomen na ischemisch letsel in vivo 6, 7 induceren. In de hippocampus, een dergelijke behandeling induceert specifiek neuronaal verlies in het CA1, maar niet in de CA3 gebied of de dentate gyrus van de hippocampus. In tegenstelling, de studie van vasculaire reacties slice cultures nu toe niet op grote schaal uitgevoerd. Een voor de hand liggende reden is het gebrek aan circulatie en bloedvat perfusie in de slice cultuur-model. Wij hebben echter eerder aangetoond dat het mogelijk is b handhavenlood schepen in CNS slice culturen voor meerdere dagen 8, 9. Het algemene doel van deze methode is om niet alleen toezicht houden op de lot van neuronen na OGD maar de studie uit te breiden tot het lot en de herinrichting van het vaatstelsel dat een belangrijk deel van de schade respons. Tot nu toe dergelijke studies het gebruik van dierproeven hebben geëist (. De Jong et al, 1999; Cavaglia et al, 2001.). In de hier gepresenteerde protocol, we detail hoe deze studies kan op entorhino-hippocampale organotypische slice cultures uitgedaagd ofwel met hypoxie of door excitotoxische laesies gevolgd door analyse van zowel neuronale overleving en bloedvaten reacties. Dit protocol is gebaseerd op een eerder gepubliceerde studie hierover 10 en kan nuttig zijn voor een laboratoriumtest vindt het neurovasculaire interacties in het CZS.

Ischemisch hersenletsel is een van de meest voorkomende en verwoestende omstandigheden afbreuk goede werking van de hersenen en leidt vaak tot blijvende functionele tekorten in de getroffen patiënten. Ondanks intensief onderzoek is er nog steeds geen effectieve behandeling optie die neuronale schade vermindert en beschermt neuronen in de ischemische gebieden van vertraagde secundaire dood. Onderzoek op dit gebied omvat gewoonlijk het gebruik van ingewikkelde en problematische diermodellen. Entorhino-hippocampus organotypische slice culturen uitgedaagd met zuurstof en glucose deprivatie (OGD) gevestigd zijn in vitro modellen die cerebrale ischemie na te bootsen. Het nieuwe aspect van deze studie is dat veranderingen in de hersenen bloedvaten bestudeerd naast neuronale veranderingen en de reactie van zowel neuronale compartiment en het vasculaire compartiment kan worden vergeleken en gecorreleerd. De in dit protocol de werkwijze van de mogelijke toepassingen van de substantieel te verbreden ofganotypic slice cultuur aanpak. De inductie van OGD of hypoxie kan met vrij eenvoudige middelen in organotypische slice cultures worden toegepast en leidt tot betrouwbare en reproduceerbare schade in neuraal weefsel. Dit staat in schril contrast met de ingewikkelde en problematische dierproeven inducerende beroerte en ischemie in vivo. Door verbreding van de analyse voor de studie van de reactie van het vaatstelsel nieuwe manieren over hoe om te behouden en te herstellen de hersenfuncties zou kunnen bieden omvatten. De slice cultuur aanpak hier gepresenteerd zou kunnen ontwikkelen tot een aantrekkelijk en belangrijk instrument voor de studie van ischemisch hersenletsel en kan nuttig zijn voor het testen van potentiële therapeutische maatregelen gericht op neuroprotection zijn.

Zuurstof glucose ontbering en hypoxie induceert neuronale dood en vermindering van bloedvaten met name in de hippocampus CA1 regio. OGD of zuurstof-deprivatie alone gedurende 15 minuten induceerde een sterke inductie van celdood zoals gezien door propidium iodide kleuring specifiek in het CA1 gebied van de hippocampus (figuur 3) hetzelfde als hiervoor 7 beschreven.
