Las neuronas son células altamente especializadas con proyecciones largas y polarizadas que dependen críticamente de transporte intracelular de las cargas esenciales para su entrega a larga distancia a diferentes microdominios axonal. Este transporte está mediado por kinesins y dyneins - dos grandes familias de proteínas motoras moleculares - que se unen a las cargas y realizar un seguimiento a lo largo de los microtúbulos polarizados en direcciones anterógrada y retrógrada, respectivamente. characterizing the composition of molecular motors on moving axonal cargo using "cargo mapping" analysis

Esta técnica supera los desafíos anteriores que utilizan métodos bioquímicos para determinar la composición media del motor de las poblaciones de vesículas purificadas a granel heterogéneos, ya que estos métodos no revelan composiciones sobre cargas móviles individuales. Además, este protocolo se puede adaptar para el análisis de otras vías de transporte y / o trata de personas en otros tipos de células para correlacionar el movimiento de las estructuras intracelulares individuales con su composición de proteínas.
