Levering van therapieën voor het centrale zenuwstelsel (CNS) in muismodellen van pediatrische ziekte blijft een uitdaging. Muizen die model pasgeboren ziektetoestanden zijn ondermaats en ontwikkelingsgebied onvolwassen en daarom moeilijk direct te injecteren in relevante structuren in het CZS. Intravasculaire injectie van therapeutische middelen is een niet-invasieve, goed verdragen methode cellen, drugs of virale vectoren leveren aan het hele lichaam inclusief het CZS 1-5 en retina 3,5-9. Vorige publicaties beschrijven temporale gezicht ader injectie met behulp van een transilluminator 10,11, zonder een dissectie microscoop 11,12, of waarbij twee personen om te injecteren 10. De in dit protocol beschreven injectietechniek is voordelig omdat een enkel individu pups kunnen injecteren, en de lichtbron om te bekijken de temporale ader wordt niet aanraken van de pup, waardoor de noodzaak voor chirurgische tape of de bevestiging van een pup op vaste ondergrondzoals een transilluminator 11. Aflevering van adeno-geassocieerde virale vector serotype 9 (AAV9) bij muizen produceert robuuste expressie in neuronen en astrocyten gehele hersenen en ruggenmerg (figuur 1). Intraveneuze injectie is een klinisch toepasbare wijze therapeutica leveren. Voor volwassen knaagdieren en grotere dieren, intraveneuze injecties zijn technisch haalbaar en routine.
