Nous démontrons un procédé de préparation de coupes de cerveau de souris en aigus physiologique au lieu de la température de la glace. Il a été démontré 11 que la qualité des tranches obtenues dans des conditions de chaleur est supérieure par rapport à ceux qui sont préparés avec des conditions de froid, à condition que la lame à trancher a un minimum de vibrations verticales. Le but de la méthode présentée est d&#39;obtenir des tranches de cerveau aigus de haute qualité pour des expériences électrophysiologiques in vitro, en particulier lors de l&#39;utilisation adulte ou même des animaux anciens. La méthode de découpage cerveau aiguë, comme décrit par Skrede et Westgaard 1 en deux phrases élégantes, est devenu l&#39;un des fondements de la recherche en neurosciences modernes et est utilisé dans d&#39;innombrables variations dans le monde entier. La qualité des tranches est reflété dans le nombre de neurones par tranche de vie, la période de temps pendant laquelle les cellules gardent leurs propriétés électrophysiologiques et morphologiques ainsi que l&#39;intégrité du tissu. De plus, la durée maximale des enregistrements stables dépend de la qualité des tranches. Ainsi, le long des décennies, la méthode de découpage original a été développé par des groupes de recherche individuels pour améliorer la récupération de la tranche après la coupe 2-10, souvent par des modifications complexes de la composition de la coupe osolutions de récupération de r (telles que l&#39;ajout d&#39;ascorbate, de la thiourée ou même H 2 O 2) et de pré-perfusion intra-cardiaque de l&#39;animal avec des solutions physiologiques refroidi. Comme il a été récemment montré 11, température physiologique pendant le découpage semble être plus bénéfique que le refroidissement de la santé neuronale; l&#39;amélioration est la plus frappante lorsque l&#39;on travaille avec des adultes (2-8 mois) les rongeurs. Eviter les variations de température considérables empêche artefacts dus à des processus dépendant de la température dans les cellules, telles que la plasticité 13 et la cinétique des canaux ioniques 13,14. De tels changements pourraient influencer la tension de la membrane et de la signalisation intracellulaire de calcium, le seuil de pic, pic et forme. Nous présentons ici un protocole de préparation des tranches de cerveau de courte durée. Cette procédure est un élément essentiel pour électrophysiologiques expériences de patch-clamp qui détermine en grande partie la qualité des résultats. Il a été démontré que l&#39;omission de l&#39;étape de refroidissement durant la procédure de coupe est bénéfique pour obtenir des tranches et des cellules saines, en particulier lorsque des structures cérébrales fortement myélinisées provenant d&#39;animaux adultes. Même si le mécanisme précis par lequel la température élevée favorise la santé de neurones ne peut être spéculé sur, il va de soi que, chaque fois que possible, la température à laquelle le découpage est effectué devrait être proche des conditions physiologiques pour éviter les artefacts liés à la température. Un autre avantage important de cette méthode est la simplicité de la procédure et donc le temps de préparation court. Dans la méthode illustrée souris adultes sont utilisés mais la même procédure peut être appliquée à des souris jeunes, ainsi que les rats. En outre, les cl de patch suivantsexpérience ampères est effectuée sur des tranches horizontales du cervelet, mais la même procédure peut également être utilisée dans d&#39;autres plans, ainsi que d&#39;autres zones postérieures du cerveau.

Tranches préparées de la manière décrite peut être utilisée pour diverses expériences électrophysiologiques et optogénétiques.
