Efter 5 timers inkubering blev cellerne høstet, vasket og farvet for T-celle delmængder. Cellerne blev derefter fikseret i PFA, permeabiliseret og farvet for interferon (IFN) -γ og analyseret ved flowcytometri.Som med andre flowcytometri-baseret assay, når cellerne behandles med fiksering og permeabilisering puffer indeholdende PFA inficerede prøver kan overføres til en BL2 laboratorium til yderligere behandling og analyse. I adskillige publicerede undersøgelser har vi anvendt ICS at måle T-celle-effektorfunktioner i WNV-inficerede mus 13,14. Selvom det godt er etableret, en væsentlig ulempe ved denne analyse er, at proceduren er meget langsommelig og kunne være mere tidskrævende, når de udføres inde BL3 faciliteter. Her blev en mikro-centrifugerør baseret ICS metode vist sig at være mere realistisk, lettere at fortsætte og mindre tidskrævende, når de udføres i en BL3 laboratorium.
