Die vorliegende Protokoll für die komplette Thymektomie eine minimal-invasive Ansatz mit einem Drei-Schicht Einschnitt Verschluss mit chirurgischen Klebstoff Anwendung entwickelt, um Komplikationen zu minimieren. Seit den frühen 1960er Jahren hat sich die Thymusdrüse für seine entscheidende Rolle in der Entwicklung der zentralen immunologische Toleranz anerkannt. Nagetier thymectomy hat sich als ein wesentlicher Vorgang bei der Definition der Rolle von Thymus der Lymphozytendifferenzierung Selbsttoleranz und Immuntoleranz in der Einstellung der Allograft Transplantation und Tumormetastasen. Die Entfernung der Ratte Thymus ermöglicht Studien mit T-Zell-Depletion oder adoptiven Transfer von definierten T-Zellpopulationen, ohne das erneute Auftreten von einheimischen naiven T-Zellen. Thymectomies bei neugeborenen Nagetieren können mit einem Saug-Technik mit zuverlässigen Ergebnissen 1 durchgeführt werden. In erwachsenen Ratten wird diese Technik mit einer ungefähren 20% Todesrate und führt häufig zu einer unvollständigen thymectomy 2. Um eine vollständige Thymektomie bei erwachsenen Ratten durchgängig zu erreichen, ist offen Belichtung des Mediastinum über eine mediane Sternotomie erforderlich. Doch diese6% 2- 4 - Verfahren ist mit Komplikationen, die Trachea Verletzungen, Blutungen und Pneumothorax, die zu einer Gesamtmortalität von 1,5 sind verbunden. In den letzten zwei Jahrzehnten Verbesserungen thymectomy Techniken perioperativen Komplikationen vermindert und wurden Überlebensraten verbessert. Intubation so dass für Überdruckbeatmung hat Pneumothorax Raten 5 verringert. Methoden der Intubation zuvor aus offenen Exposition beschriebenen Bereich um die Luftröhre zu weniger invasiven Methoden mit direkter Stimmband Visualisierung. Komplikationen im Zusammenhang mit der Intubation Verfahren verbunden sind Luftröhrenverletzung, Stimmbandbruch, unbeabsichtigte Speiseröhre Intubation und Blutungen aus Herzpunktion oder Zerstörung von der oberen Hohlvene resultieren. Außerdem kann die Nähe der unteren thymic Keulen pleuralen Auskleidung Pneumothorax führen. Hier beschreiben wir eine TechnikThymektomie durch eine minimal-invasive 2 cm Hautschnitt nach einer einfachen Methode der Intubation mit einem stumpfen Ende Angiokatheter und transkutane Luftröhrenbeleuchtung. Die thymectomy beinhaltet eine 1,5 cm Sternotomie und einen dreischichtigen Verschluß mit chirurgischen Leimauftrag auf das Mediastinum abzudichten und die Inzidenz von Blutungen und respiratorische Komplikationen zu minimieren. Dieses Verfahren zuverlässig entsteht lücken thymectomy wie durch das Verschwinden der CD4 + und CD8 + T-Zellen naive folgenden thymectomy und Abwesenheit von Thymusgewebe auf IHC Färbung nachgewiesen. Operative Zeiten und peri-prozedurale Sterblichkeit sind auf ein Minimum reduziert.

Thymektomie bei neugeborenen Nagern ist ein etabliertes und zuverlässiges Verfahren für immunologische Untersuchungen. Doch bei erwachsenen Ratten, Komplikationen von Blutungen und Pneumothorax von pleural Unterbrechung kann zu einer signifikanten Sterblichkeit führen. Dieses Protokoll ist ein einfaches Verfahren zur Ratte thymectomy, die einen Mini-Sternotomie Intubation verwendet. Intubation ist mit einer nicht-invasiv und leicht reproduzierbares Verfahren erfolgen und ermöglicht positive Druckventilation Pneumothorax zu verhindern und eine gesteuerte Luftwege, die ausreichende Zeit für eine sorgfältige Dissektion Thymus pleurale Minimierung von Störungen ermöglicht. Ein 1,5 cm Brustbeineinschnitt nimmt Kontakt mit mediastinale Gefäße und Rippenfell, während immer noch eine vollständige Visualisierung des Thymus. Nach der Belichtung des Mediastinums wird der Thymus stumpf unter Vergrößerung entfernt. Pleuraraum wird dann durch Nahtverschluss der vorge tracheale Muskeln, gefolgt von der Anwendung des chirurgischen Klebstoff abgedichtet. DieBrustkorb wird dann durch Nahtverschluss des Brustbeins, gefolgt von Nahtverschluss der Haut verschlossen. Alle thymectomies abgeschlossen waren, wie durch immunhistochemische (IHC) Färbung der mediastinalen Gewebe und Abwesenheit von naiven T-Zellen, nachgewiesen durch Durchflusszytometrie, und das Verfahren war eine 96% ige Überlebensrate. Diese Methode eignet sich, wenn Sie fertig Thymektomie mit minimalen Komplikationen für weitere immunologische Untersuchungen in athymischen erwachsenen Ratten gewünscht.

Dieses Verfahren wurde zur Erwachsenen Lewis-Ratten (n = 26) durchgeführt. Die mittlere Operationszeit betrug 15 ± 3 min. Es gab keine intraoperative Mortalität. Die mittlere Intubation Zeit - von Platzierung von Ratten auf der Intubation Gerät zur Gründung der Belüftung - betrug 45 ± 5 sec. 24 Ratten hatten eine vollständige Genesung von der Operation ohne Anzeichen von Atembeschwerden oder Blutungen durch postoperativen Tag (POD) 14.
