La thérapie génique a longtemps été suggérée comme un traitement potentiel pour la perte auditive génétique, mais le succès dans ce domaine est resté inaccessible 1. Transfert de matériel génétique exogène dans l&#39;oreille interne des mammifères en utilisant des vecteurs viraux a été étudié au cours de la dernière décennie et est en train de devenir une technique prometteuse pour le traitement des formes à la fois génétiques et acquis de la perte auditive neuf. La cochlée est potentiellement une cible idéale pour la thérapie génique pour plusieurs raisons: 1) son petit volume nécessite un nombre limité de virus nécessaire; 2) son isolement relatif des autres effets des limites des systèmes d&#39;organes secondaires; et 3) les chambres remplies de liquide faciliter viraleenvoi dans toute le labyrinthe 10, 11,12,13,14, 15. Des modèles murins de la surdité congénitale permettent une utilisation de plusieurs méthodes d&#39;étude pour suivre le développement de l&#39;oreille interne d&#39;une manière reproductible systématique. Alors que la petite taille de cochlée de souris ne présente une certaine difficulté chirurgicale, la souris sert un modèle extrêmement important dans l&#39;étude de la perte auditive génétique, avec plusieurs avantages expérimentaux sur les autres espèces 16. Des modèles de souris permettent d&#39;évaluer un éventail de caractéristiques à travers l&#39;analyse de liaison génétique, la collecte d&#39;observations morphologiques détaillées, et la simulation de scénarios pathogènes; en tant que tels, ils sont de bons candidats pour la thérapie génique à médiation virale. De vastes études génétiques chez les souris combinées avec les avancées technologiques ont permis de générer des souris génétiquement modifiées de façon reproductible dans les laboratoires 17,18, 19, 20,21. AILLEURSe, il existe de nombreux modèles à la fois pour acquises et héréditaires auditives phénotypes de perte chez la souris, ce qui permet des tests rigoureux dans ce modèle animal 22, 23,24. Ainsi, la correction auditive en utilisant la thérapie génique virale médiation dans un modèle de souris est une première étape appropriée dans la recherche d&#39;un remède pour la maladie humaine. Nous avons précédemment montré que les souris transgéniques dépourvues de glutamate transporteur vésiculaire 3 (VGLUT3) naissent sourds en raison du manque de la libération de glutamate à l&#39;IHC ruban synapse 25. Étant donné que cette mutation ne conduit pas à une dégénérescence primaire des cellules ciliées sensorielles, ces souris mutantes sont potentiellement un excellent modèle pour tester dans laquelle la thérapie génique pour cochléaire surdité congénitale. A ce jour, un certain nombre de techniques de délivrance viraux pour la thérapie génique cochléaire ont été décrites, y compris autour de diffusion de la membrane de fenêtre, l&#39;injection de la membrane de fenêtre ronde et la livraison via une cochléostomie. Il ya puissanteial avantages et les inconvénients de chacune de ces approches neuf. Nous rapportons ici une méthode chirurgicale pour la livraison de gènes à médiation virale de l&#39;oreille interne VGLUT3 KO souris à travers la membrane de la fenêtre ronde (MLE). La méthode d&#39;injection RWM post-auriculaire est peu invasive avec une excellente conservation de l&#39;audience, et est relativement rapide. Comme nous l&#39;avons déjà publié, dans un effort pour restaurer l&#39;audition dans ce modèle de souris, un vecteur AAV1 portant le gène de VGLUT3 (AAV1-VGLUT3) a été introduit dans la cochlée de ces souris sourds au jour postnatal 12 (P! @), Résultant en la restauration de l&#39;audition 26. Audience chez les souris KO VGLUT3 a été vérifiée par la réponse du tronc cérébral (ABR), tandis que l&#39;expression de la protéine transgénique a été vérifiée par immunofluorescence (IF). Cette méthodologie démontre ainsi que la thérapie génique à médiation virale peut corriger un défaut génétique qui serait sinon résultats de la surdité.

surgical method for virally mediated gene delivery to the mouse inner ear through the round window membrane

La thérapie génique, utilisé pour réaliser la récupération fonctionnelle de surdité neurosensorielle, promet d&#39;accorder une meilleure compréhension des mécanismes moléculaires et génétiques sous-jacents qui contribuent à la perte d&#39;audition. Introduction de vecteurs dans l&#39;oreille interne doit être fait d&#39;une manière qui distribue largement l&#39;agent tout au long de la cochlée, tout en minimisant les blessures aux structures existantes. Ce manuscrit décrit une approche chirurgicale post-auriculaire qui peut être utilisé pour la thérapie cochléaire de la souris en utilisant moléculaire, pharmacologique, et la livraison virale à des souris de jour postnatal 10 ans et plus par l&#39;intermédiaire de la membrane de la fenêtre ronde (MLE). Cette approche chirurgicale permet la livraison rapide et directe dans la rampe tympanique tout en minimisant la perte de sang et d&#39;éviter la mortalité animale. Cette technique consiste négligeable ou pas de dommages aux structures essentielles de l&#39;oreille interne et moyenne ainsi que les muscles du cou tout en préservant totalement audience. Pour démontrer l&#39;efficacité de cette technique chirurgicale, la glutam vésiculairetransporteur mangé trois KO (VGLUT3 KO) chez la souris sera utilisé comme un exemple d&#39;un modèle de souris de la surdité congénitale qui récupère entendu après la livraison de VGLUT3 à l&#39;oreille interne en utilisant un virus adéno-associé (AAV-1).

Pour vérifier les caractéristiques techniques et l&#39;utilité de l&#39;approche post-auriculaire pour la thérapie moléculaire cochléaire, AAV1-VGLUT3, AAV1-GFP et AAV2-GFP ont été livrés à des souris P10-12 oreille interne via le MLE. Cette approche démontre l&#39;expression du transgène avec succès dans les cellules ciliées internes (IHC) (VGLUT3 Figure 1 et la GFP Figure 2 et la GFP figure 3A), les cellules ciliées externes (CCE) (GFP Figure 2)...
